文心兰OnERS1全长基因克隆及表达分析

【摘要】:为了了解乙烯信号受体在文心兰切花衰老中的作用,利用RACE技术克隆了文心兰乙烯受体基因OnERS1(Gen Bank登录号为KP410729)。生物信息学分析表明,OnERS1基因编码631个氨基酸,分子量为70 773.7,等电点为6.82,是跨膜蛋白,与其它植物的ETR乙烯受体蛋白的相似性在74%~99%之间,属ETR1类乙烯受体。基因表达分析结果表明,OnERS1基因在根和蕊柱中表达量最高,在叶片、侧瓣和花萼中次之,在茎和唇瓣中表达量最低;文心兰切花衰老期间OnERS1基因的表达量在第0~6 d持续增加,第6 d后突然下降,第8 d后下降到一个极低的水平。与此同时,花器官中的乙烯释放从第8 d之前缓慢上升,第8~10 d急剧升高。OnERS1基因的时空表达特异性表达说明它可能与花衰老有关。
[Abstract]:In order to understand the role of ethylene signal receptor in senescence, OnERS1 (GenBank accession number KP410729) was cloned by race technique. Bioinformatics analysis showed that OnERS1 gene encodes 631 amino acids with a molecular weight of 70773.7 and a isoelectric point of 6.82. It is a transmembrane protein. The similarity of ETR ethylene receptor protein with other plants is between 74% and 99%, belonging to ETR1 type ethylene receptor. The results of gene expression analysis showed that OnERS1 gene expression was the highest in root and column, followed by leaf, lateral valve and calyx, and the lowest in stem and lip, and the expression of OnERS1 gene continued to increase on the 6th day after senescence. On the 6th day, it suddenly decreased, and on the 8th day it dropped to a very low level. At the same time, ethylene release in flower organs increased slowly from the 8th day, and increased sharply on the 8th day. The spatio-temporal expression of OnERS1 gene indicated that it might be related to flower senescence.
