基于CSSL的大白菜抽薹相关QTL上位性分析

【摘要】:抽薹性状是白菜的重要农艺性状之一。上位性效应是由非等位基因间相互作用产生的,也是复杂数量性状重要的组份,同时对白菜的抽薹性状具有重要影响。本研究以筛选的5个染色体单片段代换系和轮回亲本'RcBr'为试材,通过半轮配法构建10个双片段聚合系为试验材料,对抽薹相关QTL间的上位性效应进行分析。研究结果如下:1.本研究以F2群体,RIL群体和CSSLs群体的抽薹相关性状的QTL定位结果为参考,筛选出14个染色体片段代换系。以初步筛选出14个染色体片段代换系为试材,结合株系的表现型和基因型,进行验证试验。最终选取5个单片段代换系对上位性互作效应进行分析。每个试验材料均携带控制抽薹相关性状的QTL位点,其中有2个QTLs位点控制抽薹指数,均位于A07染色体上;其余3个QTLs位点控制开花时间,分别位于A02、A07和A08染色体上。2.本研究以欧洲白菜型油菜'RcBr'为受体亲本构建的带有抽薹相关性状QTL的5个单片段代换系为试验材料,经两两杂交,通过分子标记辅助选择(Marker-assisted selection,MAS),筛选出10个不同亲本组合的双片段聚合系。聚合系WD3(16swd001/16swd007):代换系16swd001携带的qFT-2-1对代换系16swd007携带的qFT-7-1表现上位性。聚合系WD4(16swd001/16swd009):代换系16swd001携带的qFT-2-1对代换系16swd009携带的qFT-8-1表现上位性。聚合系WD10(16swd007/16swd009):将聚合系WD10的开花时间与两亲本16swd007和16swd009的开花时间进行比较,发现开花时间未达到统计学显著差异。因此,可以判断两亲本携带的2个QTLs位点之间没有相互的叠加效应,但无法判断是否存在互作效应。3.与本研究相同亲本构建的RIL群体抽薹上位性效应鉴定结果比较,两个试验均检测到控制抽薹相关性状QTL位点之间存在上位性效应,但是检测到存在上位性效应的位点不同。本研究共检测出2对控制开花时间的QTLs位点之间存在上位性效应,分别是 qFT-2-1 与 qFT-7-1 和 qFT-2-1 与 qFT-8-1。
[Abstract]:As one of the important agronomic traits of Chinese cabbage, the epistatic effect is produced by the interaction of non-alleles and is also an important component of complex quantitative traits. At the same time, it has an important effect on the characters of Chinese cabbage. In this study, five chromosome single fragment substitution lines and their recurrent parents, RcBr', were selected as the test materials. Ten two-segment polymerization lines were constructed by half-wheel matching method to analyze the epistatic effect among QTL. The results are as follows: 1. In this study, F2 population was used. For reference, 14 chromosome fragments were selected from RIL population and CSSLs population, and 14 chromosome fragment substitution lines were selected as test materials. Finally, five single fragment substitution lines were selected to analyze the epistatic interaction effect. Each of the tested materials carried the QTL locus which controlled the characteristics of coating. Among them, 2 QTLs loci controlled the sprouting index, all of which were located on chromosome A07. The other three QTLs loci controlled blooming time, respectively, at A02. Chromosomes A07 and A08. In this study, five single-fragment substitution lines with QTL, which were derived from Brassica campestris L. as recipient parent, were used as experimental materials. After pairwise hybridization, Marker-assisted selection was selected by molecular marker-assisted selection (MAS). Ten two-fragment aggregates of different parent combinations were screened. The aggregation line WD3A16swd001 / 16swd007). : the qFT-2-1 carried by the substitution line 16swd001 is epistatic to the qFT-7-1 carried by the substitution line 16swd007. 16swd001 / 16SWd009). : the qFT-2-1 carried by the substitution line 16swd001 is epistatic to the qFT-8-1 carried by the substitution line 16swd009. 16swd007 / 16swd009: the blooming time of WD10 was compared with that of two parents 16swd007 and 16swd009. It was found that the blooming time was not statistically significant. Therefore, it could be judged that there was no superposition effect between the two QTLs loci carried by the two parents. But it is impossible to judge whether there is interaction effect. 3. Compared with the result of epistatic effect of RIL population constructed by the same parents in this study. The epistatic effect was detected between QTL loci of the traits related to the control of bolting in both experiments. However, different loci with epistatic effect were detected. In this study, two pairs of QTLs loci controlling blooming time were detected to have epistatic effect. They are qFT-2-1 and qFT-7-1 and qFT-2-1 and qFT-8-1, respectively.
