王不留行提取物对母兔哺乳性能的影响

【摘要】:本课题旨在探究向泌乳期母兔日粮中加入不同剂量的王不留行提取物对母兔泌乳及哺乳性能的影响,为王不留行黄酮苷在实际生产中的应用提供相应的理论基础。试验选择150只、体况相近、仔数一致的健康新西兰经产哺乳母兔(每胎产活仔数为10-12只),分三个周期,每个周期选择50只母乳随机分为5组(对照组和4个试验组),每个组10只母兔,试验组分别按1、2、4和8g/d的剂量在日粮中加入含有王不留行提取物(分别含王不留行黄酮苷0.004、0.008、0.016和0.032g/d),从分娩第1天开始添加,直至28日断奶,空白组的日粮和饲喂条件与试验组相同。在哺乳期间观测并记录下仔兔体重的变化、仔兔腹泻率和断奶存活率的情况、母兔血清中部分相关激素的变化、母乳中乳糖和乳铁蛋白的变化,在第28天断奶时随机抽取母兔解剖采集乳腺样本对相关基因的表达量和RNA/DNA比值进行测定,制作乳腺组织石蜡切片观测各组腺泡情况。试验结果:(1)日粮中添加王不留行提取物对断奶时仔兔生长性能的影响。母兔日粮中王不留行黄酮苷的添加对仔兔断奶时的体重有显著的影响,其中4g组和8g组显著高于空白组和1g组(P0.05);仔兔哺乳期的腹泻率受到显著影响,2g组、4g组和8g组仔兔的腹泻率低于空白组;4g组和8g组仔兔的断奶存活率高于空白组。(2)日粮中添加王不留行提取物对哺乳期母兔血清中部分激素和母乳质量的影响。日粮中添加王不留行黄酮苷对母兔血清中雌激素的含量无显著影响(P0.05);王不留行黄酮苷的添加可减缓母兔血清中PRL的含量下降趋势,极显著的增加PRL的含量,各试验组PRL的含量均显著或极显著高于空白组(P0.05,P0.01);GH含量下降的趋势可得到减缓,极显著的增加GH的含量,各试验组GH的含量均显著或极显著高于空白组(P0.05,P0.01)。日粮中添加王不留行黄酮苷可使母兔母乳中乳铁蛋白的含量在泌乳前期(第5至第9天)显著或极显著高于空白组(P0.05,P0.01),并在泌乳末期保持显著差异(P0.05);乳中乳糖的含量也会受到显著变化,泌乳的中后期(第13至第25天)试验组的乳糖含量可极显著高于空白组(P0.01),直至断奶时试验组的乳糖含量仍可显著高于空白组(P0.05)。(3)日粮中添加王不留行提取物对母兔乳腺的影响。泌乳期母兔日粮中添加一定量的王不留行黄酮苷对LACE-1、IGF-1和EGF的基因表达量有不同程度的影响。王不留行黄酮苷的添加可以显著提高LACE-1和IGF-1基因的表达量(P0.05),可极显著的影响EGF基因的表达量(P0.01)。王不留行黄酮苷的添加对乳腺组织中RNA/DNA比值有显著的降低(P0.05)。从乳腺组织的切片中可以看出王不留行黄酮苷的添加可以增加乳腺组织在断奶时仍具有泌乳活性的腺泡的数量。综上所述,向泌乳期母兔的日粮中添加王不留行提取物可以显著提高仔兔断奶时的体重,降低腹泻率,增加断奶存活率,并能提高母兔血清中PRL和GH的含量,提高母乳中乳铁蛋白和乳糖的含量,增加对LACE-1、IGF-1和EGF的基因表达量,降低RNA/DNA比值,增加乳腺中具有泌乳活性的腺泡的数量。实验结果表明,在添加量为4 g/d王不留行提取物(含王不留行黄酮苷0.016 g/d)时具有最佳的生产性能。
[Abstract]:The purpose of this study is to explore the influence of cowherb seed extracts with different doses to lactation mother rabbit diets on lactation and lactation performance of female rabbit king, without leaving the line in the application of flavonoid glycosides in actual production to provide the corresponding theoretical foundation. The added dietary flavonoid glycosides can make the content of lactoferrin in human milk in female rabbits the lactation period (fifth to ninth days) was significantly higher than the control group (P0.05, P0.01), and significant difference in late lactation (P0.05); lactose content in milk will also be significantly changed in the late lactation (thirteenth to twenty-fifth days) the test group of lactose The content can be significantly higher than the control group (P0.01), until weaning the lactose content in experimental group is still significantly higher than the control group (P0.05). (3) effects of Vaccaria segetalis extract in diet on rabbit breast. Vaccarin glycosides by adding a certain amount of lactation mother rabbit diet on LACE-1, IGF-1 and EGF the amount of gene expression have different effects. Vaccarin glycosides added increased expression of LACE-1 and IGF-1 gene (P0.05), can influence the expression of EGF gene significantly (P0.01). The flavone glycosides of RNA/DNA added milk gland tissue ratio decreased significantly (P0.05). Breast tissue slices can be seen in the number of add Vaccaria flavone glycosides can increase the breast tissue is still active at weaning milk gland. In summary, Wang added without leaving the line to extract diet during lactation in female rabbits In order to improve the weaning weight, reduce the diarrhea rate, increase the survival rate of weaning, and can increase the content of PRL and GH in serum of female rabbits, increase the content of lactoferrin and lactose in breast milk, increase of LACE-1, expression of IGF-1 and EGF gene, reduce the ratio of RNA/DNA, the number has increased the activity of breast milk in the acini. The experimental results show that the addition of 4 g/d (including vaccarin cowherb seed extracts by 0.016 g/d) has the best manufacturing performance.
