亚砷酸钠体外对细粒棘球蚴原头节生长及抗氧化酶活性的影响

【摘要】:目的研究不同浓度的亚砷酸钠(NaAsO2)对体外培养的细粒棘球蚴原头节生长及相关抗氧化酶活性的影响,探讨NaAsO2可能的作用机制。方法将不同浓度的NaAsO2(3、6、9、12和15μmol/L)作用于体外培养的细粒棘球蚴原头节,经0.1%伊红染色后于光镜下观察原头节的活力,实验独立重复3次。不同浓度NaAsO2作用原头节24h后,采用半胱氨酸天冬氨酸蛋白酶-3(caspase-3)检测试剂盒检测caspase-3酶活性;采用化学比色法或ELISA法测定NaAsO2作用后原头节抗氧化酶活性的变化。结果 12μmol/L和15μmol/L NaAsO2对原头节杀伤作用明显,从作用第1d开始,原头节活力开始下降,培养至第10d时12μmol/L组原头节存活率仅为12.64%,15μmol/L组原头节全部死亡。不同浓度(≥6μmol/L)NaAsO2作用原头节24h后caspase-3表达较正常对照组显著增高(P0.05)。NaAsO2作用后的原头节抗氧化酶(SOD、GST、HO-1和NQO-1)活性均显著下降(P0.05),且呈时间及剂量依赖性。结论亚砷酸钠能显著抑制细粒棘球蚴原头节生长,并显著降低抗氧化酶的活性,推测这种抑制作用与下调抗氧化酶活力有关,然而关于亚砷酸钠的具体作用机制有待于进一步研究。
[Abstract]:Objective to study the effects of different concentrations of sodium arsenite (NaAsO2) on the growth of proganglion and the activities of antioxidant enzymes in cultured echinococcus granulosus in vitro, and to explore the possible mechanism of NaAsO2. Methods different concentrations of NaAsO2 (3Cephalococcus granulosus 12 and 15 渭 mol/L) were applied to cultured echinococcus granulosus in vitro. After 0.1% eosin staining, the activity of the protocephalic ganglion was observed under light microscope, and the experiment was repeated three times independently. The activity of caspase-3 was detected by cysteine aspartate protease 3 (caspase-3) assay kit 24 hours after treatment with different concentrations of NaAsO2, and the changes of antioxidant enzyme activity of NaAsO2 were measured by chemical colorimetry or ELISA method. Results the killing effect of 12 渭 mol/L and 15 渭 mol/L NaAsO2 on the protocephalic ganglia was obvious. From the 1st day, the activity of the protocephalic ganglia began to decrease, and the survival rate of the 12 渭 mol/L group was only 12.64% and the survival rate of the 15 渭 mol/L group was 12.64% on the 10th day.
