板青颗粒急性与亚慢性毒性试验研究

【摘要】:为了评价板青颗粒的安全性,进行了小鼠的急性毒性试验与大鼠的亚慢性毒性试验,采用简化机率单位法测得小鼠最大耐受量(MTD)高于180 g/(kg·d),半数致死量(LD50)无法确定。进行亚慢性试验时将SD大鼠分为4组,每组20只,空白对照组饮服纯净水,板青颗粒低、中、高剂量组分别给予3,6,12 g/(kg·d)的板青颗粒于饮水中,连续28 d,分别检测体重、脏器指数、血常规指标、血液生化指标和组织切片观察。结果表明:与空白对照组比较,板青颗粒显著提高大鼠的脾脏指数(P0.05)、中性粒细胞百分比(Gran%)、血细胞比容(HCT)、红细胞体积分布宽度(RDW)(P0.05),显著降低胸腺指数、淋巴细胞百分比(Lymph%)、细胞血红蛋白浓度(MCHC)以及总蛋白(TP)、葡萄糖(GLU)、尿素氮(BUN)含量(P0.05);心脏、肝脏、脾脏、肺脏、肾脏组织切片无显著性病理变化。说明板青颗粒毒副作用和不良反应小,是安全性较高的中药方剂。
[Abstract]:In order to evaluate the safety of Banqing granules, acute toxicity test in mice and subchronic toxicity test in rats were carried out. The maximum tolerance dose (MTD) of mice measured by simplified probabilistic unit method was higher than 180 g / kg 路d). SD rats were divided into 4 groups with 20 rats in each group. The blank control group was treated with pure water, Banqing granules were low, medium and high dose groups were given 3 respectively. The body weight, organ index and blood routine were measured for 28 days.
