乌鸡源芽孢杆菌与中草药配伍对乌鸡生长性能与免疫性能的影响

【摘要】:益生芽孢杆菌作为抗生素替代品的研究很多,但关于其与复方中草药配伍的研究却很少,尤其是应用于乌鸡的研究几乎没有。本试验以珍贵药禽——乌鸡为试验对象,研究乌鸡源芽孢杆菌与中草药配伍对乌鸡生长与免疫性能的影响,旨在为新型抗生素替代品——乌鸡源芽孢杆菌与中草药配伍应用于乌鸡养殖提供理论依据。本试验从健康乌鸡胃肠道筛选到2株芽孢杆菌WJ1、WJR2,分别对其进行群体形态与个体形态观察、生理生化试验等,并以16S r DNA序列分析构建系统发育树,确定其系统发育地位:分离株WJ1判定为蜡样芽胞杆菌,分离株WJR2判定为枯草芽孢杆菌。对分离到的WJ1、WJR2进行产酶特性(淀粉酶、蛋白酶)、耐受性(人工胃液、人工肠液、猪胆盐),以及培养特性研究,确定其均为高产酶与高耐受菌株;其培养学特性为:最适p H均为6,最适生长温度均为37℃左右,其中菌株WJ1温度生长范围稍广;生长曲线特征为:两菌均在2h~12h达到对数期,WJ1的生长稳定期在12h~34h;WJR2的生长稳定期在12h~24h。将WJ1、WJR2分别与0.5g、1g、1.5g三个不同剂量的复方中草药(由党参、黄芪、茯苓等按照一定比例组成)配伍进行动物益生性试验,结果显示:不同配伍均能够不同程度提高乌鸡日增重,降低料重比,促进脾脏、法氏囊免疫器官的生长发育、保持鸡新城疫血凝抑制抗体在较高水平,还能有效的提高乌鸡体内免疫球G蛋白(Ig G)的含量。其中试验组Ⅳ(WJ1与1g复方中草药配伍)在日增重、提高脾脏指数、维持ND效价在较高水平效果最为明显。与对照组相比,日增重最多可达23.6%(P0.05);脾脏免疫指数42d时最高达29.1%(P0.05),ND效价最高达60%(P0.05)。试验组Ⅵ(WJR2与0.5g复方中草药配伍)料重比降低最为明显,36-42d最多可降低19.1%(P0.05)。试验组Ⅷ(WJR2与1.5g复方中草药配伍)提高法氏囊免疫指数、IgG含量效果最为明显。与对照组相比,法氏囊免疫指数35d时最高达50.5%(P0.05);Ig G含量28d最高为50.5%(P0.05)。
[Abstract]:Bacillus probioticus as a substitute for antibiotics has been studied a lot, but few studies have been done on its compatibility with compound Chinese herbal medicine, especially on silky chicken. To study the effect of compatibility of Bacillus spp and Chinese herbal medicine on growth and immune performance of silky chicken. The aim of this study was to provide a theoretical basis for the application of a new antibiotic substitute, Bacillus from silky chicken, in combination with Chinese herbal medicine in silky chicken culture. In this study, two strains of Bacillus sp. WJ1 and WJR2 were screened from gastrointestinal tract of healthy silky chicken, and their population morphology was studied. And individual morphological observation, The phylogenetic tree was constructed by 16s r DNA sequence analysis. The phylogenetic status of the phylogenetic tree was determined. The isolated strain WJ1 was identified as Bacillus cereus. The isolated strain WJR2 was identified as Bacillus subtilis. The enzyme producing characteristics (amylase, protease, tolerance (artificial gastric juice, artificial intestinal fluid, porcine bile salt), and culture characteristics of the isolated WJ1 / WJR2 were determined to be high enzyme and high tolerance strains.
