基于高通量测序研究铜、汞和铅对鸡盲肠微生物多样性的影响

【摘要】:为探讨重金属对海兰褐蛋鸡盲肠结构和微生物多样性的影响。实验选取30日龄海兰褐蛋鸡72羽,随机分成4组:对照组(CK)、铜组(Cu)、汞组(Hg)和铅组(Pb),每组3个重复,每个重复6羽。CK组饲喂纯净水,Cu组饲喂含量为150mg/L CuCl2纯净水,Hg组为含量140mg/L HgCl2纯净水,Pb组为含量350 mg/L PbCl2纯净水。于实验第30d、60d、90d采取鸡盲肠及盲肠内容物。观察鸡盲肠组织结构和应用Illlumina-Hiseq 250高通量测序技术分析微生物群落结构和多样性的变化。结果如下:1、CK组盲肠组织学结构完整,黏膜分布规律,隐窝处腺体发育正常。与对照组相比,Cu组、Hg组有少量黏膜脱落并伴有少量淋巴细胞增生,Pb组肠黏膜大量断裂脱落并伴有大量淋巴细胞增生。2、72羽海兰褐蛋鸡盲肠微生物共得到5527565个有效序列,2431个OTUs,分类属于28个门,348个属。3、共享核心菌门主要为厚壁菌门、拟杆菌门、疣微菌门、变形菌门和柔膜菌门。属水平以拟杆菌属比例最高,占19.20%,其次是钩藤菌属。4、Cu组后壁菌门,乳酸菌及粪球菌等优势菌门及菌属丰度最高。5、Hg组柔膜菌门,魏氏梭菌属和丹毒丝菌属等致病菌丰度最高。6、Pb组拟杆菌门、后壁菌门等优势菌门丰度降低;魏氏梭菌属、丹毒丝菌属、普雷沃氏菌属等致病菌属丰度升高。结果说明,重金属对鸡盲肠组织有一定的损伤作用,并影响肠道菌群的结构及多样性。
[Abstract]:In order to study the effects of heavy metals on the cecal structure and microbial diversity of Hailan brown laying hens, 72 30 day-old Hayland brown laying hens were randomly divided into four groups: control group, copper group, mercury group (Hgg) and lead group (3 replicates in each group). Each repeated 6 plume. CK group was fed with pure water containing 150 mg / L CuCl2 pure water and Hg group with a concentration of 140 mg / L HgCl2 pure water pure water Pb group was fed with 350 mg/L PbCl2 pure water. The contents of chicken caecum and cecum were taken on the 30th day of the experiment, and the contents of cecum and cecum were observed in the chicken caecum group. The changes of microbial community structure and diversity were analyzed by high-throughput sequencing technique of Illlumina-Hiseq 250. The results were as follows: the structure of cecum was intact in the 1: 1 CK group. Mucosal distribution, Compared with the control group, there was a small amount of mucosal exfoliation and a small amount of lymphocyte proliferation. The intestinal mucosa in Pb group was ruptured and exfoliated with a large number of lymphocyte proliferation. A total of 5527565 active sequences were obtained, including 2431 OTUs, belonging to 348 genera of 28 phylum. Bacteroid phylum, Microphylum verrucum, Proteus and Fleuromycetes. The highest proportion of Bacteroides was found in genus 19.20, followed by phylum of posterior wall in group of Leptacterium, Lactobacillus and faecal coccus, and the highest abundance of Bacteroides in the group of 5.5Hg, the second was the phylum of the posterior wall of the genus Leptacterium, the other dominant bacteria such as lactobacillus and faecococcus. The abundance of pathogenic bacteria such as Clostridium Weissii and Rhizoma erysipelas is the highest. The abundance of Bacteroides phylum and phylum of the posterior wall is decreased, the abundance of pathogenic bacteria such as Clostridium Weissii, Staphylococcus erysipelis, Genus Pravo is increased, the results show that, Heavy metals can damage the cecum tissues and affect the structure and diversity of intestinal flora.
