川金果榄油膏治疗大鼠烫伤的药效学观察

【摘要】:烧伤是指由各种物理或化学因素,包括热液、蒸汽、火焰、电能、化学物质、放射线等所造成组织损伤的病症。针对频发的烧伤,中药治疗的研究也在不断深入。本课题主要观察川金果榄油膏对大鼠烫伤创面的保护作用,并进一步探讨其作用机理,为促进川金果榄油膏的临床应用及新剂型开发提供坚实的理论基础。主要实验内容及结果如下:1大鼠浅Ⅱ度烫伤模型的建立大鼠背部脱毛24h后,采用沸水加热的100g砝码分别作用于大鼠皮肤5s、10s、15s和20s,于烫伤24h后切取烫伤创面全层皮肤,制备病理切片,观察皮肤表皮层及真皮层的病理变化。得出结论:烫伤5s能建立稳定的大鼠浅II度烫伤模型,烫伤10s、15s、20s能建立不同程度的大鼠深II度烫伤模型。2川金果榄油膏对大鼠烫伤创面的保护作用SD大鼠20只,雌雄各半,随机分成5组,每组4只。伤后30min分别于各组烫伤部位给药,以均匀覆盖创面为宜,灭菌纱布包扎,每天给药1次,连续给药至创面完全愈合,模型组给予油膏基质处理。以上皮完全覆盖作为伤口完全愈合的依据。与模型组相比,高剂量组(P0.01)和中剂量组(P0.05)均能明显缩短创面的愈合时间。在伤后第7d,与模型组相比,高剂量组和中剂量组愈合率显著升高(P0.05),伤后14d亦延续这种趋势。从整个愈合过程中各组烫伤大鼠的病理组织学切片来看,川金果榄油膏能在早期减轻炎症细胞的浸润,加快创面纤维细胞和毛细血管生长,后期加快表皮覆盖,促进创面愈合。3川金果榄油膏促进大鼠烫伤创面愈合的作用机理SD大鼠84只,雌雄各半,其中正常组4只,其余大鼠随机均分为5组,每组16只。烫伤后,各组分别给予药物治疗,于伤后1d、3d、7d、14d各组随机选取4只大鼠,雌雄各半,尾静脉采血离心得血清,切取创面组织标本,保存于-80℃冰箱。通过对实验大鼠烫伤后1d-14d的部分生理指标的测定发现:伤后第3天,各组烫伤大鼠血清中IL-6含量上升趋势明显并达峰值,且均与正常组差异极显著(P0.01),间接反映造模成功。从创面组织中SOD和MDA含量可以看出,烫伤创面均在早期有一个不断加深的过程。而药物组创面羟脯氨酸含量均在伤后第3天达峰,早于模型组,且达峰浓度明显高于模型组。伤后第7天,与模型组相比,川金果榄油膏低剂量组创面组织含水量显著下降(P0.05),伤后第14天效果更加明显(P0.01);川金果榄油膏高、中、低剂量组SOD活性均与模型组差异极显著(P0.01),各组MDA含量也均保持下降趋势,高剂量组下降幅度大,与模型组差异明显(P0.05);各药物组创面组织中EGF含量的达峰时间均早于模型组,且在伤后第7天,川金果榄油膏高、中剂量组创面组织中EGF的含量显著高于模型组(P0.05);各组烫伤大鼠血清中IL-6含量随时间延长而下降,其中,与模型组相比,川金果榄油膏高剂量组(P0.01)和中剂量组(P0.05)下降趋势更明显;各组血清中TGF-p的含量在伤后第7天达峰,且与模型组相比,高剂量组烫伤大鼠血清中TGF-p的含量显著升高(P0.01)。结论:1.运用加热5分钟的100g砝码致伤5s能建立稳定的大鼠浅II度烫伤模型。2.川金果榄油膏能较好地提高烫伤不同时间点创面的愈合率,缩短烫伤创面的愈合时间,对烫伤创面显示出较好的修复作用。3.川金果榄油膏促进烫伤创面愈合的作用机制可能涉及促进创面胶原生成,抑制脂质过氧化反应,促进细胞增殖和表皮爬行,控制部分炎症介质的过度合成和释放,促进烫伤大鼠EGF和TGF-p含量的升高。
[Abstract]:Burn is a disease of tissue damage caused by various physical or chemical factors, including hydrothermal, steam, flame, electric energy, chemical material, radiating line and so on. The study of Chinese medicine treatment for frequent burns is also in depth. This subject is mainly to observe the protective effect of the ointment on scald wounds in rats and further explore its effect. Mechanism to provide a solid theoretical basis for promoting the clinical application of Chuanjin olive oil ointment and the development of new dosage forms. The main experimental contents and results are as follows: 1 rats with shallow second degree scald model of 1 rats were established by boiling water heated 100g weights to 5S, 10s, 15s and 20s respectively, and scalded after 24h were cut to scald after scalding 24h. All layers of skin, pathological sections were prepared, and the pathological changes of skin epidermis and dermis were observed. Conclusion: scald 5S can establish a stable II degree scald model in rats, scald 10s, 15s, 20s can establish different degree of rat deep II scald model.2 Sichuan golden fruit olivan ointment to protect the scald wound of rats, SD rats 20, male and female. The infiltration of the disease cells, quickening the growth of fibroblast and capillary in the wound, accelerating the skin cover in the later period, promoting the healing of the wound healing.3 and promoting the healing of scald wound in rats, 84 rats and male and male 4 rats were divided into 5 groups randomly, and the other rats were randomly divided into 5 groups, 16 rats in each group. The peak value was significantly different from that of the normal group (P0.01), which indirectly reflected the success of the mold making. From the content of SOD and MDA in the wound tissue, the burn wound had a continuous deepening process in the early stage. The content of hydroxyproline in the wound surface of the drug group were all in the third Tianda peak in the post injury, earlier than the model group, and the peak concentration was significantly higher than that in the model group. The peak time of EGF content in the wound tissue of each drug group was earlier than that in the model group, and the content of EGF in the wound tissue in the middle dose group was higher than that in the model group (P0.05) in the middle dose group seventh days after the injury, and the IL-6 content in the serum of the scalded rats decreased with the time prolonging, which was compared with the model group. The decreasing trend of the high dose group (P0.01) and the medium dose group (P0.05) was more obvious, and the content of TGF-p in the serum of each group was seventh Tianda peak after injury, and compared with the model group, the content of TGF-p in the serum of scalded rats increased significantly (P0.01). It can promote cell proliferation and epidermal crawling, control excessive synthesis and release of some inflammatory mediators, and promote the increase of EGF and TGF-p contents in scalded rats.
