山羊精液液态保存过程中氧化还原态的变化及人工授精

【摘要】:精液的长期保存通常采用冷冻保存,该技术在牛上已实现了商业化,但是山羊的精液冷冻效果却不理想,受精率和妊娠率较低,因此主要以液态保存为主。研究中发现,山羊精液液态保存至12天与保存16天时活率均能达到40%左右,但利用保存12天和16天的精液人工授精母羊的受胎率却出现显著差异,有必要进一步研究和分析母羊的受胎率差异的原因,为进一步提高山羊精液液态保存方案奠定基础。本实验利用mZAP的改进液MES稀释液,添加20%的卵黄包被,然后再用MES稀释液保存精子,研究不同保存时间精子活力和存活率、精子质膜和顶体完整性、精液稀释液中氧化还原状态的变化以及对保存12天和15天的精液进行人工授精,结果如下:(1)精子的活力和存活率随着保存时间的延长而逐渐下降,其中活力在第14天下降幅度最大,存活率在第15天显著降低;(2)精子质膜完整性在第14天显著下降,顶体完整性从第15开始有显著下降;(3)精液稀释液中的氧化还原状态随保存时间的延长也发生变化,TOS含量逐渐升高,第14天变化幅度最大;还原态也是随着保存时间的延长而升高,但从第14天开始,还原态开始下降。第14-16天精子的OSI的上升幅度达到最大;(4)保存12天的精液人工授精后受胎率与对照组无显著差异,但保存15天组受胎率显著降低。上述结果表明,山羊精液液态保存过程中,精子活力、存活率、质膜完整、顶体完整都会随保存时间的延长而逐渐下降,保存12天的精液具有与对照组几乎相当的受胎率,但保存15天精液受胎率明显下降。
[Abstract]:Cryopreservation is usually used in the long-term preservation of semen, which has been commercialized on cattle. However, the freezing effect of goat semen is not ideal, the fertilization rate and pregnancy rate are low, so liquid storage is the main method. It was found that the survival rate of goat semen stored in liquid state for 12 days and 16 days could reach about 40%, but the pregnancy rate of artificial insemination ewes with 12 and 16 days of preservation was significantly different. It is necessary to further study and analyze the reasons for the difference of conception rate of ewe, so as to lay a foundation for further improving the plan of liquid preservation of goat semen. In this experiment, the modified MES diluent of mZAP was added with 20% egg yolk coating, then the sperm was preserved with MES diluent. The sperm motility and survival rate, the integrity of plasma membrane and acrosome were studied. The changes of redox state in semen diluent and artificial insemination of semen stored for 12 and 15 days were as follows: (1) the viability and survival rate of spermatozoa decreased with the prolongation of preservation time. The motility of spermatozoa decreased significantly on the 14th day and the survival rate decreased significantly on the 15th day. (2) the sperm plasma membrane integrity decreased significantly on the 14th day, and the acrosomal integrity decreased significantly from the 15th day. (3) the redox state of semen dilute also changed with the prolongation of storage time, and the content of TOS increased gradually, and the biggest change was on the 14th day, and the redox state also increased with the prolongation of preservation time, but from the 14th day, the content of TOS increased. The reduced state began to decrease. (4) there was no significant difference between the pregnancy rate of the semen after 12 days of preservation and the control group, but the pregnancy rate of the 15 days group was significantly lower than that of the control group. The results showed that the sperm motility, survival rate, plasma membrane integrity and acrosome integrity decreased with the prolongation of preservation time during liquid preservation of goat semen, and the semen stored for 12 days had almost the same conception rate as the control group. However, the pregnancy rate of semen decreased significantly after 15 days of preservation.
