利用PCR方法对延边地区猪布氏杆菌病的流行病学调查

【摘要】:本课题的研究目的是利用PCR方法对延边地区不同县市总计320头尚未注射预防布氏杆菌病疫苗的猪进行检测,对猪布病的流行情况进行流行病学调查,从而为延边地区猪布病的防控提供科学参考。在研究过程中,对320头抽检猪首先使用琥红平板凝集检测方法进行初步检测,再使用试管凝集方法进一步检测,最终利用PCR方法对被检猪进行猪布病流行病学调查,以统计延边地区不同县市猪布病的流行情况。试验检测结果如下:1.RBPT检测阳性率约为31.3%(100/320),阴性率约为68.7%(220/320);和龙市阳性率最高为36.7%(22/60),敦化市阳性率最低为20.0%(6/30)。2.SAT检测阳性率约为21.3(68/320),阴性率约为78.7%(252/320);和龙市阳性率最高为26.7%(16/60),延吉市阳性率最低为10.0%(2/20)。3.PCR检测阳性率约为13.8%(44/320),阴性率约为86.2%(276/320);和龙市阳性率最高为16.7%(10/50),延吉市阳性率最低为5.0%(1/20)。4.抽检200只圈养猪,RBPT检测阳性率为32.0%(64/200),SAT检测阳性率为25.0%(50/200),PCR检测阳性率为 17.0%(34/200)。5.抽检120只散养猪,RBPT检测阳性率为30.0%(36/120),SAT检测阳性率为15.0%(18/120),PCR 检测阳性率为 8.3%(10/120)。6.夏季抽检的198只猪中,和龙市、敦化市检测阳性率相同为22.2%,汪清县为18.8%,图们市、龙井市以及安图县阳性率相同为16.7%,珲春市为12.5%,延吉市为8.3%(1/12);198头份猪全血PCR检测总阳性率为17.2%(34/198)。7.冬季抽检的122只猪中,龙井市检测阳性率为14.3%,图们市、安图县阳性率相同为12.5%,汪清县为11.1%,和龙市为8.3%,延吉市、敦化市、珲春市阳性率相同为0;61头份猪全血PCR检测总阳性率为8.2%(10/122)。8.利用PCR方法对延边地区部分猪场320头猪进行布病流行病学调查后,可以确定延边地区是猪布病的流行地区,且和龙市流行最严重,而且夏季阳性率高于冬季,圈养阳性率高于散养。
[Abstract]:The purpose of this study was to investigate the prevalence of brucellosis by using PCR method in 320 pigs from different counties and cities in Yanbian area which had not been vaccinated against brucellosis. So as to provide scientific reference for prevention and control of pig cloth disease in Yanbian area. In the course of the study, 320 pig samples were detected by using the method of hunoon-red plate agglutination first, and then further detected by the method of test tube agglutination. Finally, the epidemiological investigation of pig brucellosis was carried out by using PCR method. The epidemic situation of pig cloth disease in different counties in Yanbian area was analyzed. The results are as follows: 1. The positive rate of RBPT is about 31.3% (100 / 320), the negative rate is about 68.7% (220 / 320), the highest positive rate is 36.7% (22 / 60) in Hulong City, the lowest positive rate is 20.0% (630 / 30) .2.SAT positive rate is about 21.3% (630 / 320) in Dunhua City, and 78.7% (252 / 320) in Helong City, 26.7% (16 / 60) in Harong City, and 26.7% (16 / 60) in Yanjin City. The lowest positive rate was 10.0% (2 / 20) .3.The positive rate of PCR was about 13.8% (44 / 320), the negative rate was about 86.2% (276 / 320), the highest positive rate of Helong was 16.7% (10 / 50), and the lowest positive rate of Yanji was 5.0% (1 / 20) .4. The positive rate of RBPT was 32.0% (64 / 200). The positive rate of SAT was 25.0% (50 / 200). The positive rate of PCR was 17.0% (34 / 200) .5. The positive rate of RBPT was 30.0% (36 / 120). The positive rate of SAT was 15.0% (18 / 120). The positive rate of PCR was 8.3% (10 / 120) .6. Among 198 pigs sampled in summer, the positive rate in Helong City and Dunhua City was the same as that in Wangqing County (18.8%), Tumen City, Longjing City and Antu County were 16.7%, Hunchun City 12.5%, Yanji City 8.3% (1 / 12) 198 whole Blood PCR positive rate 17.2% (34 / 198) .7. Of the 122 pigs sampled in winter, the positive rate in Longjing City was 14.33.The positive rate in Tumen and Antu counties was 12.5. the positive rate was 11.1cm in Wangqing County, 8.3in Helong city, 8.3in Yanji city, Dunhua city and Hunchun city. The total positive rate of whole blood PCR test was 8.2% (10 / 122) .8. Based on the epidemiological investigation of brucellosis in some pig farms in Yanbian area by PCR method, it can be concluded that Yanbian area is the endemic area of swine brucellosis, and the epidemic is the most serious in Helong city, and the positive rate in summer is higher than that in winter. The positive rate in captivity was higher than that in rest.
