蛹虫草多糖对鸡新城疫疫苗免疫效果的影响

【摘要】:为研究蛹虫草多糖(CMP)对鸡新城疫苗免疫效果的影响,将200只14日龄雏鸡随机分为5组,除空白对照组(BC)外,均接种新城疫疫苗,28日龄第二次接种。第一次接种疫苗的同时,三个CMP40组鸡分别肌肉注射低、中、高浓度的多糖,免疫对照组(VC)和BC组注射等量生理盐水,每天1次,连续3 d。在第1次接种后的第7(D7)、14(D14)、21(D21)、28(D28)、35(D35)和42(D42)天采血,分别用MTT法和血凝抑制(HI)试验测定外周血淋巴细胞增殖和血清抗体效价变化,在D14、D21和D28用ELISA测定血清IL-4和IFN-γ浓度。结果显示,CMP40L组在所有时间点的A570值和血清抗体效价显著高于VC组和BC组(P0.05);低剂量的CMP40发挥佐剂性能的效果最好,同时可促进机体对IFN-γ和IL-4的分泌。
[Abstract]:In order to study the effect of Cordyceps militaris polysaccharide (CMP) on the immune effect of chicken Newcastle vaccine, 200 14-day-old chicks were randomly divided into 5 groups, all of them were inoculated with Newcastle disease vaccine for the second time at 28 days except the blank control group (BC). At the same time, the three CMP40 groups were injected with low, medium and high concentrations of polysaccharides intramuscularly, while the control group, (VC) and BC groups, were injected with the same amount of normal saline once a day for 3 days. Blood samples were collected on the 7th (D7) 14 (D14) 21 (D21) 28 (D28) -35 (D35) and 42 (D42) days after the first inoculation. The proliferation of peripheral blood lymphocytes and serum antibody titers were measured by MTT assay and hemagglutination inhibition (HI) test. Serum IL-4 and IFN- 纬 concentrations were measured by ELISA at D14D21 and D28, respectively. The results showed that the A570 value and serum antibody titer in CMP 40L group were significantly higher than those in VC group and BC group at all time points (P0.05). Low dose CMP40 had the best effect on adjuvant performance and could promote the secretion of IFN- 纬 and IL-4.
