葡萄籽提取物减缓西杂肉牛空肠上皮细胞热应激反应的功能与机制研究

【摘要】:葡萄籽提取物(Grape seed extract,GSE)是多酚类物质的重要来源之一,具有抗氧化、抗炎和免疫调节的功能。本试验旨在研究GSE缓解肉牛空肠上皮细胞(Jejunum epithelial cells,JEC)热应激反应的作用与机制。在肉牛(西门塔尔♂×秦川牛Q的杂交后代)空肠上皮细胞培养基中分别添加0、10、20、40和80μg/mL的GSE,然后将各浓度梯度下的JEC细胞分别在37℃、40℃、43℃和46℃下进行2h的培养。研究显示,当细胞培养温度为40℃时,添加GSE时细胞谷胱甘肽过氧化物酶(GSH-Px)活性(P=0.04)、细胞活力(P=0.004)、细胞表皮生长因子(Epidermal growth factor,EGF;P=0.03)和细胞表皮生长因于受体(Epidermal growth factor receptor,EGFR;P=0.01)mRNA表达均有升高。在同一温度下,添加GSE降低细胞中自细胞介素(Interleukin 8.IL-8)(P=0.01)、肿瘤坏死因子α(TNF-α)mRNA的表达,同时降低IL-1β(P=0.02)、Toll样受体4(TLR4;P=0.04)蛋白和热休克蛋白70(HSP70;P0.001)的浓度。当细胞培养温度为43℃时,添加GSE组细胞的过氧化氢酶活性(P=0.04)、细胞活力(P0.001).EGF(P0.001)和EGFR(P0.001)mRNA的表达均得到提升,而IL-1β(P0.001).TLR4(P=0.03)和HSP70(P0.001)的浓度以及IL-8(P0.001)、TLR4(P=0.002)和TNF-α(P0.001)的mRNA表达却显著降低。T×GSE互作效应在JEC细胞IL-1β(P0.001)、IL-8(P0.001)、TNF-α(P=0.01)、HSP70(P=0.04)的浓度和细胞活力(P0.001)上得到体现。本试验研究结果表明,在特定温度范围内,GSE能够从提高细胞抗氧化能力以及降低炎性反应两个方面缓解JEC细胞的热应激反应。
