腺胃型传染性支气管炎病毒的分离鉴定

【摘要】:从河北病鸡肿大腺胃中分离到1株病毒(命名为CK/CH/HBHS/07F株),经鸡胚传代、血凝特性测定、新城疫病毒(Newcastle disease virus,NDV)干扰试验、传染性支气管炎病毒(Infectious bronchitis virus,IBV)膜蛋白(membrane protein,M)基因序列分析、人工感染试验等方法对该病毒进行了鉴定。该分离毒株感染鸡胚的尿囊液没有直接血凝活性,经胰酶处理后可凝集鸡红细胞,其血凝活性可被IBV阳性血清所抑制;经鸡胚传至第3代时,可致死鸡胚或出现侏儒胚;序列分析结果表明,该病毒M基因与18株不同致病型IBV毒株间的核苷酸和氨基酸序列同源性分别为82.8%~94.1%、82.4%~97.4%,与腺胃型毒株CK/CH/LJL/04I同源性最高,该分离株能人工复制出与自然病例相似的病理变化。初步证实CK/CH/HBHS/07F株为腺胃型IBV。
[Abstract]:A strain of virus (named CK/CH/HBHS/07F strain) was isolated from the stomach of the enlarged gland of Hebei disease chicken. After the passage of chicken embryo, the determination of hemagglutination characteristics, and the interference test of Newcastle disease virus (Newcastle disease virus,NDV), (Infectious bronchitis virus, of infectious bronchitis virus was obtained. IBV) membrane protein (membrane protein,M) gene sequence analysis, artificial infection test and other methods to identify the virus. The hemagglutination activity of allantoic fluid of chicken embryo infected by this isolate strain was not direct hemagglutination activity, and the hemagglutination activity of chicken erythrocyte could be agglutinated after trypsin treatment, and its hemagglutination activity could be inhibited by IBV positive serum, and it could kill chicken embryo or produce dwarf embryo at the third generation of chicken embryo. Sequence analysis showed that the nucleotide and amino acid sequence homology between M gene of the virus and 18 strains of different pathogenic IBV strains were 82.8% and 82.4%, respectively. The CK/CH/LJL/04I strain had the highest homology with the adenogastric virus strain, and the isolated strain could replicate the pathological changes similar to those of the natural cases. Preliminary confirmation of CK/CH/HBHS/07F strain as adenogastric IBV.
