日粮钴添加水平对生长獭兔生产性能、氮代谢性能和毛囊发育的影响

【摘要】:本课题研究日粮不同钴添加水平对生长獭兔生产性能、代谢性能和毛囊发育的影响。试验选取基线水平一致的生长獭兔200只,随机分为5组,分别饲喂不同钴添加水平(0、0.1、0.4、1.6、6.4mg/kg)的试验日粮,采用单因素试验设计方法研究其对生长獭兔的影响。试验结果如下:(1)在初始体重(IBW)无显著差异的前提下,日粮钴添加水平对试验末期体重(EBW)有显著影响(P0.05),对平均日采食量(ADI)(P0.01)有极显著影响。对平均日增重(ADG)、料重比(F/G)、全净膛率、半净膛率、肉品质、胸腺指数、脾指数、肝指数、肠绒毛高度、隐窝深度、V/C均无显著影响(P0.05),随着日粮钴添加水平的升高,EBW、ADI、半净膛率、胸腺指数、十二指肠绒毛高度均先升高后降低,并且都在0.4 mg/kg时最高。(2)日粮钴添加水平对粪氮(FN)、可消化氮(DN)、DN/食入氮(IN)、沉积氮(RN)/DN、总蛋白和白蛋白均无显著影响(P0.05),对IN、UN、RN、RN/IN有显著影响(P0.05),并且随着日粮钴添加水平的升高,IN、RN和RN/IN先降低后升高,UN显著降低,在钴添加水平为0.4~1.6 mg/kg时达到最大值。日粮钴添加水平对胰蛋白酶活有显著影响(P0.05),在钴添加水平为1.6 mg/kg时最高。日粮钴添加水平对血清尿素含量有显著影响(P0.05),随着日粮钴添加水平的升高,血清尿素先降低后升高,在钴添加水平为1.6 mg/kg时最低。日粮钴添加水平对粪、尿、肌肉、脾脏和肾脏中钴含量有显著影响(P0.05),并且随着日粮钴添加水平的升高,钴含量均显著增加。(3)日粮钴添加水平对獭兔的皮张面积、皮张重量、皮张厚度没有显著影响(P0.05)。随着日粮钴添加水平的升高,皮张重量呈现先增加后降低的趋势,且在钴添加水平为0.4mg/kg时最高。日粮钴添加水平对獭兔的被毛长度、毛囊密度有显著性影响(P0.05),随着日粮钴添加水平的升高,毛囊密度呈现先增加后降低的趋势,钴添加水平为6.4mg/kg时毛囊密度最低可能是因为钴添加量太高导致轻微中毒。分别选取钴添加量为0和6.4 mg/kg的组进行荧光定量分析和Western Blotting技术分析,结果发现日粮钴添加水平对Tyrosinase、BMP2、BMP4有显著性影响(P0.05),并且高钴日粮组比低钴日粮组表达水平都升高,表明Tyrosinase、BMP2、BMP4参与了钴对毛囊的调控,同时AMPK、Wntlob信号通路没有参与钴对毛囊的调控。综合本试验测定指标,生长獭兔日粮钴适宜添加水平为0.4 mg/kg(实际日粮中钴含量为0.60 mg/kg),其中钴添加量为6.4mg/kg时獭兔有轻微中毒症状。
