AA肉鸡肝脏生物活化AFB1的关键Ⅰ相代谢酶及姜黄素的干预机理

  黄曲霉毒素B1(AFB1)是已知的黄曲霉毒素中研究最深也是毒性最强的一类霉菌次级代谢产物。肉鸡摄入经AFB1污染的饲料,通常会引起中毒,降低肉鸡的免疫力,并造成严重的经济损失。经证实,肝脏是蓄积和代谢AFB1的主要器官,AFB1经肝脏代谢产生的有毒代谢产物残留在肉鸡体内,可以造成肉鸡肝损伤甚至癌症;残留在肉鸡组织中的AFB1的代谢物通过食物链可被人类摄入,从而产生公共卫生危害。目前,关于肉鸡体内的CYP2A6在AFB1致肉鸡亚慢性肝损伤中所扮演的角色尚不明确。姜黄素是从姜黄根茎中提取的多酚类化合物,具有抗炎、抗氧化、抗肿瘤等作用。近期研究表明,姜黄素在脂肪肝综合征、肝炎、肝硬化和肝癌等肝脏疾病中起着保护肝脏的作用。姜黄素能否通过影响细胞色素酶p450表达尤其是CYP2A6对AFB1诱导AA肉鸡亚慢性肝损伤发挥保护作用尚不清楚。研究表明,AFB1属于前致癌物,须通过肉鸡肝脏的细胞色素酶p450的生物活化后才能发挥其毒性和致癌作用,且代谢酶存在种属的差异。已有文献报道证明,人类肝脏中CYP2A6以及鼠源的CYP2A5是同源物,分别为人或鼠体内催化AFB1代谢活化为若干种致癌物质(毒物)的C...
