【摘要】: 目的建立CXCL16(结合磷脂酰丝氨酸和氧化型脂蛋白的清道夫受体)转基因小鼠模型,并观察其血脂水平的变化。 方法构建带Flag分子标签的真核表达载体CXCL16-trans,显微注射入C57BL"6J小鼠受精卵原核,导入假孕母鼠输卵管后培养子代小鼠。通过PCR法及Western Blot鉴定小鼠基因型,获得CXCL16转基因小鼠模型(CXCL16 transgenic mouse ,CXCL16 trans )。8周龄F5代CXCL16 trans鼠和野生型小鼠(C57BL"6J)各10只,经12周高脂饲料喂养后,应用紫外分光仪法检测喂养前后小鼠的总胆固醇、甘油三脂、低密度脂蛋白及高密度脂蛋白的水平。 结果1.构建携带分子标签Flag的表达载体CXCLl16-trans质粒经酶切与测序,鉴定序列正确。在171只原代小鼠中,有26只转基因阳性Founder小鼠。目前F5代71只小鼠中,有6系共31只转基因阳性小鼠。 2.CXCL16 trans与野生型小鼠经12周高脂饲料喂养后,前者血清胆固醇、甘油三酯及低密度脂蛋白水平明显高于后者,而血清高密度脂蛋白水平低于后者。 结论1.成功建立携带分子标签Flag的CXCL16转基因小鼠模型。 2.高脂饲料喂养的CXCL16转基因小鼠,其血清总胆固醇、甘油三酯及低密度脂蛋白明显增高,而血清高密度脂蛋白水平明显降低,提示基因CXCL16在动脉粥样硬化的发生发展过程中可能起着促进的作用。
[Abstract]:Objective to establish a CXCL16 transgenic mouse model with phosphatidyl serine and oxidized lipoprotein scavenger receptor, and to observe the changes of lipid levels in CXCL16 transgenic mice. Methods Eukaryotic expression vector CXCL16-transcoding with Flag molecular label was constructed and microinjected into prokaryotic of C57BL "6J mouse fertilized egg." The progeny mice were cultured in the fallopian tubes of pseudopregnant mice. The genotypes of the mice were identified by PCR and Western Blot. CXCL16 transgenic mouse model CXCL16 transgenic mouse was obtained. There were 10 CXCL16 trans mice of F5 generation and 10 C57BL "6J" mice of CXCL16 trans at the age of 8 weeks. The mice were fed with high fat diet for 12 weeks. The levels of total cholesterol, triglyceride, low density lipoprotein and high density lipoprotein in mice before and after feeding were detected by ultraviolet spectrometer. The expression vector CXCLl16-trans plasmid carrying molecular label Flag was constructed by restriction endonuclease digestion and sequencing. The sequence was correct in 171 primary mice. 2. There were 26 transgenic Founder mice and 31 transgenic mice in 6 lines out of 71 F5 generation mice. 2. The levels of serum cholesterol, triglyceride and low density lipoprotein in CXCL16 trans and wild-type mice were significantly higher than those in wild type mice. The serum HDL level was lower than that of the latter. The model of CXCL16 transgenic mice carrying molecular label Flag was successfully established. 2. 2.The serum total cholesterol, triglyceride and low density lipoprotein of CXCL16 transgenic mice fed with high fat diet were significantly increased, while the serum high density lipoprotein level was obviously decreased. It is suggested that gene CXCL16 may play a promoting role in the development of atherosclerosis.
