【摘要】: 实验目的:探讨骨髓基质干细胞标记失踪的合理有效方法。 实验方法:以兔骨髓基质干细胞(rabbit marrow stromal stem cells)为研究对象,抽取兔髂脊骨髓血,按贴壁细胞分离法分离骨髓基质干细胞MSCs,进行体外扩增培养。用293FT细胞和PWPXLd、PAX2、PMD2.G三质粒系统包装携带绿色荧光蛋白(EGFP)基因的慢病毒(lentivirus),收获慢病毒后,用病毒侵染兔骨髓基质干细胞,在荧光显微镜下观察转染结果。 结果:分离培养出兔骨髓来源的MSCs,并用携带绿色荧光蛋白( EGFP)基因的慢病毒成功感染兔骨髓MSCs。 结论:获得兔骨髓MSCs并进行慢病毒载体介导的基因修饰,为基质干细胞基因工程和组织工程研究奠定了基础。
[Abstract]:Objective : To investigate the rational and effective method of marker disappearance of bone marrow stromal cells . Methods : The rabbit bone marrow stromal cells ( MSCs ) were isolated from rabbit bone marrow stromal cells . MSCs were isolated from bone marrow stromal cells by adherent cell separation method . The MSCs were isolated from bone marrow stromal cells by adherent cell separation method . The lentivirus ( lentivirus ) carrying green fluorescent protein ( EGFP ) gene was packed with 293FT cells and PWPXLd , PAX2 and PMD 2.G three plasmid systems . The rabbit bone marrow stromal cells were infected with virus , and the transfection results were observed under fluorescence microscope . Results : MSCs from rabbit bone marrow were isolated and cultured , and rabbit bone marrow MSCs were successfully infected with lentivirus carrying green fluorescent protein ( EGFP ) gene . Conclusion : To obtain rabbit bone marrow MSCs and carry out the gene modification mediated by the slow virus vector , a foundation is laid for the research of gene engineering and tissue engineering of stromal cells .
