【摘要】: 目的:观察intermedin (IMD)对大鼠肾脏缺血再灌注损伤(IRI)中氧化应激作用的影响。 方法:健康雄性Wistar大鼠30只随机分为假手术组、IRI组、转空质粒组、转肾上腺髓质素(ADM)质粒组、转IMD质粒组。动物右肾切除后,用超声微泡技术将质粒转染入肾脏,1周后制作肾脏IRI模型。半定量分析肾脏病理损伤,比色法检测肾组织超氧化物岐化酶(SOD)、髓过氧化物酶(MPO)及caspase-3活性、脂质过氧化物丙二醛(MDA)含量,免疫组织化学方法检测细胞间粘附分子-1 (ICAM-1)、P-选择素及内皮素1(ET-1)表达,原位末端标记法(TUNEL)检测肾组织细胞凋亡。 结果:1、手术组大鼠发生急性肾小管坏死,以IRI组病理损伤最重,转染IMD后病理损伤明显减轻。2、IRI组SOD活性明显降低,MDA含量明显增加;MPO活性明显增高,ICAM-1、P-选择素表达明显增加;ET-1表达明显增加;caspase-3活性明显增高,大量肾小管上皮细胞凋亡。转染IMD后SOD活性明显增高,MDA的生成明显减少;MPO活性明显降低,ICAM-1、P-选择素表达明显减少;ET-1表达明显减少;caspase-3活性明显降低,肾小管上皮细胞凋亡减轻,与转ADM组之间没有明显差别。转空质粒组和IRI组以上指标没有明显差别。 结论:IMD在肾脏局部高表达对肾脏IRI有保护作用,其机制至少部分与抑制氧自由基生成、炎细胞浸润及炎症因子ICAM-1、P-选择素生成,ET-1生成,细胞凋亡有关,从而减轻肾组织局部氧化应激反应产生的活性氧族。
[Abstract]:Objective : To observe the effect of intermedin ( IMD ) on oxidative stress in rats with renal ischemia - reperfusion injury ( IRI ) . Methods : 30 healthy male Wistar rats were randomly divided into sham operation group , IRI group , empty plasmid group , adrenomedullin ( ADM ) plasmid group and transferred IMD plasmid group . After the animal right kidney was removed , the plasmid was transfected into the kidney for 1 week . The expression of superoxide dismutase ( SOD ) , myeloperoxidase ( MPO ) , caspase - 3 activity , lipid peroxide malondialdehyde ( MDA ) content and immunohistochemical method were used to detect the expression of intercellular adhesion molecule - 1 ( ICAM - 1 ) , P - selectin and endothelin - 1 ( ET - 1 ) . TUNEL was used to detect the apoptosis of renal tissue cells . Results : 1 . The acute tubular necrosis of renal tubular epithelial cells occurred in the operation group . The expression of SOD in IRI group was significantly decreased , the content of MDA was significantly increased , the expression of ICAM - 1 and P - selectin was significantly increased , the expression of ICAM - 1 and P - selectin was significantly decreased , the expression of ICAM - 1 and P - selectin was significantly decreased , the expression of ICAM - 1 and P - selectin was significantly decreased , the expression of ICAM - 1 and P - selectin was significantly decreased , the expression of ICAM - 1 and P - selectin was significantly decreased , and no significant difference was found between the expression of caspase - 3 and apoptosis of renal tubular epithelial cells . Conclusion : The local high expression of IMD has protective effect on renal IRI , and its mechanism is at least partially related to the inhibition of oxygen free radical formation , inflammatory cell infiltration and inflammatory factor ICAM - 1 , P - selectin formation , ET - 1 formation and apoptosis , thus reducing reactive oxygen species produced by local oxidative stress reaction in renal tissue .
