【摘要】: 血管内皮祖细胞(endothelial progenitor cells,EPCs)是指具有特异性归巢于血管新生组织并能分化增殖为成熟内皮细胞的一群干/祖细胞,是中胚层卵黄囊血岛原血干细胞(hemangioblast)向内皮细胞分化过程中的一个过渡阶段,该细胞在出生后的新生血管发生/生成中起重要作用。本实验从既简化实验步骤又可以获得足量内皮祖细胞的角度出发,通过6%羟乙基淀粉(Hydroxyethyl Starch, HES)沉降法和Percoll非连续性密度梯度离心法联合应用,分离、培养脐静脉血中单个核细胞,在VEGF、bFGF、BPE等因子的体外诱导下,单个核细胞分化成为内皮祖细胞,利用免疫组织化学、免疫荧光、流式细胞仪等技术,从形态学、表面标志以及功能特性等多角度逐渐深入地研究单个核细胞成功的分化成为内皮祖细胞,并且进一步向内皮细胞分化的过程。本实验为进一步体内移植内皮祖细胞,研究其在视网膜新生血管中的作用机制及寻找有效的防治措施提供了细胞来源。
[Abstract]:Endothelial progenitor cells (endothelial progenitor cells, EPCs) is a specific homing to angiogenesis and tissue differentiation into mature endothelial cells of a group of stem / progenitor cells, mesoderm is the yolk sac blood Shimabara blood stem cells (Hemangioblast) into endothelial cells in a transitional stage in the process of neovascularization in the cells in the postnatal / play an important role in the generation of this experiment. From the experimental procedure is simplified and can get sufficient endothelial progenitor cells through the perspective of 6% hydroxyethyl starch (Hydroxyethyl, Starch, HES) Percoll non precipitation method and combined application of continuous density gradient centrifugation and cultured mononuclear cells, umbilical vein the blood in VEGF, bFGF, BPE and other factors induced in vitro, differentiation of mononuclear cells into endothelial progenitor cells by immunohistochemistry, immunofluorescence, flow cytometry, from morphology, Surface markers and functional properties such as multi angle gradually in-depth study on differentiation of mononuclear cells into endothelial progenitor cells successfully, and further differentiation into endothelial cells. This experiment for further in vivo transplantation of endothelial progenitor cells, cell sources for research and find out the effective control measures in retinal neovascularization mechanism.
