促性腺激素预处理的小鼠排卵过程中ADAMTS-1在卵巢中的表达的研究

【摘要】: 目的:探讨促性腺激素预处理的小鼠排卵过程中ADAMTS-1在卵巢中的表达与排卵的关系。 方法:将40只22-24天大小的C57/BL6雌性小鼠随机分成10组,分别为孕马血清促性腺激素(PMSG)组(D1组)、注射hCG后2h、4h、8h、12h、16h、24h、48h、72h(D2组-D9组)及对照组生理盐水组(D0组),每组4只小鼠。采用原位杂交进行组织学定位,逆转录聚合酶链反应(RT-PCR)进行半定量,在mRNA水平上检测排卵过程中ADAMTS-1在小鼠卵巢中的动态表达。 结果: ADAMTS-1主要表达于卵巢表面大卵泡的颗粒细胞,而在卵巢中央的小卵泡和闭锁卵泡没有表达。对照组(D0组)、PMSG组(D1组)、注射hCG后48 h和72 h(D8组、D9组)均无目的基因ADAMTS-1的表达,注射hCG后2h、4h、8h、12h、16h、24h(D2组-D7组)均有目的基因ADAMTS-1的表达,且从注射hCG后2 h至12 h卵巢ADAMTS-1mRNA的表达逐渐增强,注射hCG后12 h(D5组)达到高峰,其ADAMTS-1mRNA表达的相对值为(1.357±0.328),与各组比较均有极显著差异( P 0.05),此后表达逐渐下降,至注射hCG后48 h(D8组)无表达,但D2、D3、D4、D6、D7组之间无显著性差异( P 0.05)。 结论: ADAMTS-1在注射hCG后12h达到高峰,此时正是促性腺激素预处理的小鼠即将排卵的时间。ADAMTS-1可能是排卵过程中重要的信号传导分子。
[Abstract]:Aim: to investigate the relationship between ADAMTS-1 expression and ovulation during ovulation of mice pretreated with gonadotropin. Methods: forty C57 / BL6 female mice aged 22 to 24 days were randomly divided into 10 groups: group D 1 (gestational horse serum gonadotropin) group and group D 1, 2 h after injection of hCG. Group D _ (9) and control group (n = 4) and normal saline group (n = 4). Histological localization was performed by in situ hybridization. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the dynamic expression of ADAMTS-1 in mouse ovary at mRNA level. Results: ADAMTS-1 was mainly expressed in granulosa cells of large follicles on the surface of ovary, but not in small follicles and atretic follicles in central ovary. In PMSG group, there was no expression of ADAMTS-1 at 48 h and 72 h after hCG injection, and the expression of ADAMTS-1 was found at 4 h after hCG injection. The expression of target gene ADAMTS-1 was observed in D _ 2 group (n = 12) and D _ 2 group (n = 24). The expression of ovarian ADAMTS-1mRNA increased gradually from 2 h to 12 h after hCG injection, and reached the peak in group D 5 12 h after hCG injection. The relative value of ADAMTS-1mRNA expression was 1.357 卤0.328, which was significantly different from that of each group (P 0.05), and then decreased gradually. There was no expression of D8 at 48 h after hCG injection, but there was no significant difference between D2D3, D4, D6 and D7 groups (P 0.05). Conclusion: ADAMTS-1 reached its peak at 12 hours after hCG injection. This is the time of ovulation in gonadotropin pretreated mice. ADAMTS-1 may be an important signal transduction molecule during ovulation.
