特异性免疫应答在真菌性角膜炎发病和转归中的作用

【摘要】: 目的普遍认为非特异性免疫应答在真菌性角膜炎(FungalKeratitis,FK)发病和转归中起着至关重要的作用。本课题将探讨实验性的FK能否诱导特异性免疫应答。方法通过角膜基质注射白色念珠菌孢子建立小鼠的FK模型,鼻吸入白色念珠菌孢子诱导真菌性肺炎,皮下注射热灭活的白色念珠菌孢子进行主动免疫。在初次或再次感染真菌性角膜炎后的不同时间点收集血浆,用ELISA测定IgA、IgG、IL4和IFNγ。用组织免疫荧光法检测受染角膜组织沉积的免疫球蛋白和浸润的淋巴细胞。结果免疫活性小鼠基质注射白色念珠菌孢子后发生典型的FK并在三周内痊愈。当再次用白色念珠菌孢子感染已痊愈的角膜时,相比于初次感染炎症更轻并愈合更快。感染真菌性肺炎痊愈的小鼠和主动免疫的小鼠对FK的抵抗力更强。相比较于首次诱导FK的小鼠,以前接触过白色念珠菌的小鼠再次诱导FK后血浆中产生更多的IgG和IgA含量,角膜中有更多的免疫球蛋白沉积和淋巴细胞浸润。分析细胞因子表明FK诱导的免疫反应偏向于Th1型。结论FK诱导Th1型特异性免疫应答和免疫记忆,先前接触过真菌制品提高宿主对其后角膜感染相同真菌的抵抗力。在高危人群中主动免疫可能是预防真菌性角膜炎的一种有效策略。
[Abstract]:The purpose is generally believed that the nonspecific immune response in fungal keratitis (FungalKeratitis, FK) incidence and outcome plays a crucial role. This paper will discuss the experimental FK can induce specific immune responses in mice. Methods FK model built by intrastromal injection of Candida albicans spores, nasal inhalation of Candida albicans induced by fungal spores the spores of Candida albicans pneumonia, subcutaneous injection of heat inactivated by active immunization. Plasma was collected at different time points of primary and secondary infection of fungal keratitis after determination of IgA, ELISA, IgG, IL4 and IFN with gamma. Immunofluorescence detection of immunoglobulin staining of corneal tissue deposition and infiltration of lymphocytes. The immune activity of mice after injection of matrix of Candida albicans spores typical FK and recovered in three weeks. When again with Candida albicans spores infection has been cured film angle When compared to the initial infection inflammation lighter and faster. The healing of fungal pneumonia in mice cured and active immunization of mice infected with resistance to FK stronger. For the first time compared to FK mice induced by IgG and IgA, the more content of plasma before contact with Candida albicans in mice induced by FK again, in the cornea immunoglobulin deposition and lymphocyte infiltration. More analysis of cytokines demonstrated that the immune response induced by FK in favor of Th1. Conclusion FK induced Th1 specific immune response and immune memory, previously exposed to fungal products improve host on subsequent corneal infection the same fungal resistance. In a high-risk population of active immunization may be a effective strategies for prevention of fungal keratitis.
