中西医结合法建立大鼠病窦模型的手术方法和病理研究

【摘要】:目的:(1)探索用介入法建立大鼠病态窦房结综合征(sick sinus syndrome, SSS,简称病窦)模型的手术方法;(2)比较甲醛、氢氧化钠、冰醋酸、无水乙醇、鸦胆子油这五种药物哪种为介入法建立大鼠病窦模型的最佳试剂;(3)通过实验中各种指标的对比研究证明用介入法建立大鼠病窦模型比开胸法更优越。 方法:先将一批健康大鼠筛查心电,选出心率在350-450次/分之间大鼠60只,然后随机分为六组,分别是甲醛开胸湿敷组、甲醛介入组、氢氧化钠介入组、冰醋酸介入组、无水乙醇介入组、鸦胆子油介入组,每组10只。甲醛开胸湿敷组:乌拉坦常规麻醉,连接呼吸机,开胸暴露窦房结,用40%甲醛棉球湿敷窦房结区;甲醛介入组:无水乙醚麻醉,先将大鼠胸壁胸骨右缘第二肋间剪一小口,将自制标测电极导管沿此小口插入胸腔,按照心电图波形来确定给药部位。40%甲醛溶液用微量注射器经自制标测电极导管局部滴注造模。其余介入组实验方法同甲醛介入组。测量HR、进行病理观察、记录术后存活率及术程并进行对比研究。 结果:(1)与开胸法比较介入法术程短、操作更简单且死亡率低;(2)各组大鼠心率均有不同程度下降,通过心率变化的统计分析,得出10%氢氧化钠为最适合的造模药物,效果稳定;(3)从病理组织学分析来看,乙醇、冰醋酸、鸦胆子油虽然对窦房结有一定破坏,但损伤较轻;甲醛和氢氧化钠对窦房结造成的损伤更接近病窦的临床病理改变。 结论:1.用介入方法建立大鼠病窦模型方法简便,定位准确,副损伤小,成功率高。2.介入方法建立大鼠病窦模型的最佳试剂是10%的氢氧化钠溶液,其建立的模型稳定性好,可以用于较长时间的药效学研究。
[Abstract]:Objective to explore the surgical method of establishing sick sinus syndrome (SSS) model of sick sinus syndrome (SSS) in rats by interventional method. (2) compare formaldehyde, sodium hydroxide, acetic acid, anhydrous ethanol and Brucea javanica oil, which is the best reagent to establish the rat model of sick sinus by interventional method; 3) the comparative study of various indexes in the experiment proved that the interventional method was superior to the open chest method in the establishment of the model of the diseased sinus in rats. Methods: a batch of healthy rats were screened and 60 rats with heart rate between 350 and 450 beats / min were selected. Then they were randomly divided into six groups: the open chest wet compress group and the formaldehyde intervention group. Sodium hydroxide intervention group, acetic acid intervention group, anhydrous ethanol intervention group, Brucea javanica oil intervention group, each group, 10 rats. Wet application of 40% formaldehyde cotton ball to sinus node area; Formaldehyde intervention group: anaesthetized with anhydrous ether, we cut the second intercostal part of the sternum of the rat chest wall first, and inserted the self-made measuring electrode catheter into the thoracic cavity along this small opening. According to the electrocardiogram waveform, the injection site of .40% formaldehyde solution was determined by micro-syringe. The experimental methods of other intervention groups were the same as those of formaldehyde intervention group and HR were measured. Pathological observation, postoperative survival rate and procedure were recorded and compared. Results compared with the open chest method, the intervention spell duration was shorter, the operation was simpler and the mortality rate was lower. (2) the heart rate of rats in each group decreased in different degrees. Through statistical analysis of heart rate changes, it was concluded that 10% sodium hydroxide was the most suitable drug for modeling, and the effect was stable. 3) from histopathological analysis, ethanol, glacial acetic acid and Brucea javanica oil damaged sinoatrial node slightly; The damage caused by formaldehyde and sodium hydroxide to the sinoatrial node is closer to the clinicopathological changes of the diseased sinus. Conclusion: 1.The method of establishing rat sick sinus model by interventional method is simple, accurate, less collateral damage and high success rate. The best reagent of interventional method is 10% sodium hydroxide solution. The established model has good stability and can be used in pharmacodynamics study for a long time.
