人类牙胚组织中SHH信号通路的表达检测

【摘要】:SHH (Sonic Hedgehog)信号通路在小鼠早期牙胚的发育过程中起着关键的作用,对于其在小鼠磨牙中的分子生物学机制人们已经做了大量的研究。为了进一步探索SHH信号通路在人类牙胚发生发育过程中所起的调控作用以及可能的分子机制,本实验率先检测了SHH信号通路中的关键基因在人类牙胚发育早期的乳门牙和乳磨牙中的表达模式,这些基因包括了:SHH, PTCl, SMO, GLI1,GLI2以及GLI3,通过比对其在小鼠牙胚相应发育时期的表达模式来进一步揭示SHH信号通路在不同物种中的潜在表达调控机制。研究表明在蕾状期以及帽状期这些基因在人类与小鼠中的表达模式十分相近,但个别基因的表达模式存在差异,比如SHH信号在人类牙胚相应时期的表达区域就不局限在釉结节中,而是在整个内釉上皮中都可以检测到其表达,而下游信号分子的表达区域范围比小鼠中略大。在人类牙齿发育的钟状期SHH、PTCl的表达模式与小鼠中完全一致,但此时无论在人类的乳门牙还是乳磨牙中均检测不到其它基因的表达。通过对本实验结果的分析我们认为SHH信号通路在人类与小鼠牙胚发育的早期表达模式总体来说十分相近,很可能SHH信号通路在人类中以相同的调控模式调节着上皮和间充质细胞的增殖,诱导早期牙齿发育,而在人类牙齿的钟状期以后,SHH信号通路的调控作用很可能别别的家族的信号分子所取代,或者与其它分子通路之间形成新的信号转导网络。
[Abstract]:SHH Sonic Hedgehog signaling pathway plays a key role in the early development of mouse tooth germ. In order to further explore the regulatory role of SHH signaling pathway in the development of human tooth germs and possible molecular machineries, a great deal of research has been done on its molecular biological mechanism in mouse molars. System. This study was the first to detect the expression patterns of key genes in the SHH signaling pathway in human dental germs in the early stages of primary incisor and primary molars. These genes include: SHH, PTCl, SMO. GLI1, GLI2 and GLI3. By comparing their expression patterns in the corresponding developmental stages of mouse tooth germs, the potential expression regulation mechanism of SHH signaling pathway in different species was further revealed. The results showed that these genes were expressed in humans in bud and cap stages. The expression pattern was very similar to that in mice. However, the expression patterns of individual genes are different. For example, the expression region of SHH signal in the corresponding period of human tooth germ is not confined to the enamel nodule, but can be detected in the whole inner enamel epithelium. The expression range of downstream signal molecules was slightly larger than that of mice, and the expression pattern of SHHPCl in the bell stage of human tooth development was completely consistent with that in mice. However, no other genes were detected in human primary incisor or primary molars at this time. Based on the analysis of the results of this study, we believe that the expression pattern of SHH signaling pathway is early in the development of human and mouse tooth germs. It's very similar in general. It is very likely that the SHH signaling pathway regulates the proliferation of epithelial and mesenchymal cells in the same manner in humans and induces early tooth development after the bell phase of human teeth. The regulation of SHH signaling pathway is likely to be replaced by other family signaling molecules or to form a new signal transduction network with other molecular pathways.
