固体培养和液体培养对铜绿假单胞菌双鞭毛影响的比较

【摘要】: 细菌鞭毛染色在细菌鉴定中是很重要的技术。由于以前的鞭毛染色方法如Ryu染色法、Leifson染色法、镀银染色法、Gray法等都存在着染色液的不稳定性,着色欠佳,操作方法复杂等缺点。2002年,谷海瀛发表镀银染色新方法,该方法具有简单、快速、重复性好、染色后鞭毛清晰等多种优点。2005年,谷海瀛、李凡发表了细菌液体膜过滤法新技术,用于鞭毛染色,以前细菌液体培养鞭毛染色都是使用离心法,染色制片菌体不易散开,聚集成团,背景不干净.膜过滤法完全克服了这些缺点,操作快速,细菌鞭毛染色效率高。该方法改变了鞭毛肉汤成份,细菌生长快速,有利于鞭毛生长发育。 1872年首次报告铜绿假单胞的发现,以前所有文献报告铜绿假单胞菌只有1根鞭毛,称为单端单鞭毛菌,直到2005年谷海瀛首次发现铜绿假单胞菌具有1-2根鞭毛,称为单端双鞭毛。这一发现是在菌种接种哥伦比亚绵羊血平板培养后鞭毛染色观察到的。 通过液体培养膜过滤技术鞭毛染色,比较固体培养和液体培养铜绿假单胞菌双鞭毛的差别,研究结果提示:铜绿假单胞菌液体培养双鞭毛细胞数与单鞭毛细胞数的比值高于固体培养。
[Abstract]:Bacterial flagellum staining is an important technique in bacterial identification because of the previous flagellum staining methods such as Ryu staining and silver plating staining. In 2002, Gu Haiying published a new method for silver staining, which is simple, rapid and reproducible. In 2005, Gu Haiying and Li Fan published a new technique of bacterial liquid membrane filtration, which was used in flagellum dyeing. The bacteria are not easy to disperse, gather into clusters, and the background is not clean. Membrane filtration method completely overcomes these shortcomings, rapid operation, high efficiency of bacterial flagellum staining. This method changes the flagellum broth composition. The rapid growth of bacteria is beneficial to the development of flagella. In 1872, the discovery of Pseudomonas aeruginosa was first reported. All previous literatures reported that Pseudomonas aeruginosa had only one flagella, known as Monoflagella monocytogenes. Until 2005, Pseudomonas aeruginosa was first found to have 1-2 flagellum, known as double flagellum, in Gu Haiying. This discovery was observed by flagellum staining after inoculated with Colombian sheep blood plate culture. The difference of double flagella between solid culture and liquid culture of Pseudomonas aeruginosa was compared by liquid culture membrane filtration technique. The results showed that the ratio of double flagellum cells to single flagella cells in liquid culture of Pseudomonas aeruginosa was higher than that in solid culture.
