促红细胞生成素对人骨髓间充质干细胞增殖和细胞周期的影响

【摘要】: 目的探讨不同剂量的促红细胞生成素(erythropoietin,EPO)对人骨髓间充质干细胞(mesenchymal stem cells,MSCs)增殖和细胞周期的影响。 方法采用Percoll淋巴细胞分离液,密度梯度离心法分离骨髓间充质干细胞;无血清条件下,以不同剂量0.25、0.50、1.00、2.00、4.00、8.00U/mL的促红细胞生成素与骨髓间充质干细胞共培养为实验组,不加促红细胞生成素的骨髓间充质干细胞为对照组;24h、48h、72h后,MTT法检测骨髓间充质干细胞的增殖情况;以10U/mL的促红细胞生成素与骨髓问充质干细胞共培养72h后,流式细胞仪检测MSCs的细胞周期。 结果本实验骨髓间充质干细胞与促红细胞生成素共培养后,骨髓间充质干细胞的增殖指数明显增加,促红细胞生成素呈时间、剂量依赖性促进骨髓间充质干细胞增殖,以4U/mL促红细胞生成素促骨髓间充质干细胞增殖作用最明显;与对照组比较,促红细胞生成素干预组G_0/G_1期比例降低,S期和G_2/M期细胞比例增加,两组差异显著(P<0.05)。 结论在促红细胞生成素培养条件下可以促进更多的骨髓间充质干细胞进入DNA合成期,从而有更多的细胞分裂,产生大量的增殖细胞,并且这种影响呈时间、剂量依赖性。
[Abstract]:Objective to investigate the effects of different doses of erythropoietin (EPO) on the proliferation and cell cycle of mesenchymal stem cells (MSCs) in human bone marrow mesenchymal stem cells (MSCs). Methods Bone marrow mesenchymal stem cells (BMSCs) were isolated by Percoll lymphocyte isolation and density gradient centrifugation, and co-cultured with erythropoietin (EPO) and bone marrow mesenchymal stem cells (BMSCs) in different doses of 0.250.50U / mL and 4.00U / mL without serum. The proliferation of bone marrow mesenchymal stem cells (BMSCs) without erythropoietin was detected by MTT assay after 24 h, 48 h and 72 h, and then cultured with 10 U / mL Erythropoietin for 72 h. The cell cycle of MSCs was detected by flow cytometry. Results after co-culture of bone marrow mesenchymal stem cells with erythropoietin, the proliferation index of bone marrow mesenchymal stem cells increased significantly, and erythropoietin promoted the proliferation of bone marrow mesenchymal stem cells in a dose-dependent manner. The proliferation of bone marrow mesenchymal stem cells (BMSCs) stimulated by 4U / mL erythropoietin was the most obvious. Compared with the control group, the proportion of G0 / G1 phase of erythropoietin intervention group decreased the proportion of S phase and G2 / M phase, the difference between the two groups was significant (P < 0.05). Conclusion under the condition of erythropoietin culture, more bone marrow mesenchymal stem cells can enter DNA synthesis stage, thus more cells divide, and a large number of proliferative cells are produced, and this effect is time and dose dependent.
