导尿管球囊扩张法构建胃食管反流病动物模型方法的改进

【摘要】:目的通过复合法,构建兼具食管下段抗反流屏障缺陷和胃排空障碍的胃食管反流病(GERD)动物模型。方法选取12只5月龄新西兰大白兔,随机分为两组(各6只),分别行球囊扩张法和复合法。建模成功后4周处死动物,行食管炎建模效果评价和胃动力学检测。结果 2组动物均未见死亡,全部建模成功。按照组织学食管炎指数标准,组间食管炎严重程度指数差异无统计学意义。食管炎建模后可见明显细胞间隙增宽,2组间差异无统计学意义。应用复合法建模对胃排空产生显著影响,较球囊扩张法胃排空率显著降低。结论本研究建立的模型既存在食管下段抗反流屏障缺陷,又存在胃肠动力异常所造成的胃排空延迟,提供了一种满足临床治疗需要的更理想模型。
[Abstract]:Objective to construct GERD animal model of gastroesophageal reflux disease (GERD) with lower esophageal anti-reflux barrier defect and gastric emptying disorder. Methods Twelve 5-month-old New Zealand white rabbits were selected. The animals were randomly divided into two groups (6 rats in each group) by balloon dilatation method and composite method. The animals were killed 4 weeks after the model was established, and the effects of esophagitis modeling and gastric dynamics were evaluated. Results No death was found in the two groups. All of them were successfully modeled. According to the histologic esophagitis index standard, There was no significant difference in the severity index of esophagitis between the two groups. There was no significant difference between the two groups after modeling esophagitis. The compound method was used to model the gastric emptying. The gastric emptying rate was significantly lower than that of balloon dilatation. Conclusion the model has not only the defect of anti-reflux barrier in lower esophagus, but also the gastric emptying delay caused by abnormal gastrointestinal motility. It provides a more ideal model to meet the needs of clinical treatment.
