碱性成纤维生长因子对脂肪干细胞分化为心肌细胞的影响

【摘要】:【目的】探讨碱性成纤维生长因子(b FGF)对脂肪干细胞(ADSC)分化为心肌细胞的影响。【方法】从SD大鼠中提取、分离、培养、鉴定ADSC后,评价ADSC分化能力。将成功建立心肌梗死(MI)模型的60只大鼠随机分4组(n=15/组):PBS组、b FGF组、ADSC组及b FGF+ADSC组。建立心梗1周后分别注射PBS、b FGF、ADSC及b FGF+ADSC,4周后用心脏超声测量心梗大鼠的左室射血分数(EF),取大鼠心脏检测左室梗死面积、微血管密度(MVD)及ADSC分化。【结果】成功分离ADSC,流式细胞分析显示大多数ADSC表达CD29和CD90,不表达CD34和CD45。ADSC能被诱导分化成成脂、成骨细胞。b FGF+ADSC组及ADSC组的EF高于PBS组和b FGF组(P0.01),b FGF+ADSC组的EF高于ADSC组(P0.01),PBS组和b FGF组的EF差别无统计学意义(P=0.33)。b FGF+ADSC组及ADSC组的梗死面积小于PBS组和b FGF组(P0.01),b FGF+ADSC组的梗死面积小于ADSC组(P0.01),PBS组和b FGF组的梗死面积差别无统计学意义(P=0.28)。四组中b FGF+ADSC组的MVD最大(P0.01)。ADSC可分化为c Tn T阳性细胞、SMA阳性细胞及fⅧ阳性细胞。【结论】b FGF可促进ADSC分化成心肌细胞和新生血管,改善心脏功能。
[Abstract]:[objective] to investigate the effect of basic fibroblast growth factor (bFGF) on the differentiation of adipose stem cells (ADSCs) into cardiomyocytes. [methods] ADSC was isolated, cultured and identified from SD rats. To evaluate the differentiation ability of ADSC, 60 rats were randomly divided into 4 groups: ADSC group, B FGF group, FGF group and b FGF ADSC group. After 1 week of myocardial infarction was established, PBSb FGFA ADSC and b FGF ADSCC were measured by echocardiography after 4 weeks of myocardial infarction. The left ventricular ejection fraction (LVEF) of rats with myocardial infarction was measured, and the infarct size of left ventricle was measured. Microvessel density (MVD) and ADSC differentiation. [results] ADSCs were successfully isolated. Flow cytometry showed that most ADSC expressed CD29 and CD90, but not CD34 and CD45.ADSC could be induced to adipose. The EF of osteoblast. B FGF ADSC group and ADSC group was higher than that of PBS group and b FGF group. The EF of FGF ADSC group was higher than that of ADSC group P0. 01 ADSC group and b FGF group. There was no significant difference in EF between b FGF ADSC group and b FGF group. The infarct area of b FGF ADSC group and ADSC group was smaller than that of PBS group and b FGF group. The infarct area of group B was smaller than that of group ADSC (P 0.01). There was no significant difference in infarct area between group B and group B FGF.
