几种因素对血管环张力及细胞钙离子的影响

【摘要】: 乳酸作为糖代谢的中间产物在外周神经系统(联系中枢神经系统与全身各器官的神经,包括脑神经和脊神经)的作用已经得到非常广泛研究。剧烈运动时,外周乳酸浓度显著增高,乳酸积累被认为是产生疲劳的机制之一。另外,近年来的一些研究发现,运动中体内氨的积累与疲劳的产生也密切相关。SO_2是常见的大气污染物,尤其在一些工业较发达、人口较密集或燃用高硫煤较多的城市或地区SO_2超标非常严重。因此,SO_2对人体健康影响的研究日益受到社会的关注。 为了从信号转导角度探讨SO_2对大鼠的降压作用机理、SO_2对血管舒张作用的机理,本实验以Wistar大鼠为实验对象,测定SO_2对体外培养平滑肌细胞、内皮细胞的影响。另外,本实验初步研究了乳酸以及氨对Wistar大鼠胸主动脉血管的舒张作用。 1、乳酸和氨对血管舒张的影响及与SO_2的联合作用 研究了乳酸和氨对血管舒张的影响以及其与其他药物的联合作用。采用文献的方法制备大鼠胸主动脉环,Medlab-U/4C501生物信号采集处理系统测定血管环张力,分别加入不同浓度的乳酸、氨水以及其他药物。结果:①在10~(-6)mol·L~(-1)NE预收缩后,加入乳酸(0.5、1、5、10、20、100mmol·L~(-1))后血管环张力逐渐减小,表现为舒张作用,且与乳酸的浓度呈剂量依赖性。分别加入10μmol·L~(-1)、100μmol·L~(-1)SO_2;10μmol·L~(-1)、100μmol·L~(-1)亚硫酸盐;3nmol·L~(-1)、5nmol·L~(-1)硝普钠,并同时加入不同浓度的乳酸后血管环张力逐渐减小,均表现为舒张作用。但其舒张比小于乳酸或其他药物单独作用后舒张比之和。②氨(0.3-20mmol·L~(-1))对大鼠主动脉环有浓度依赖性的舒张作用;低浓度氨(0.3-1.0mmol·L~(-1))对血管的舒张作用无明显影响,而高浓度氨(2.0-20 mmol·L~(-1))的影响比较明显。 2、SO_2对血管细胞产生Ca~(2+)浓度的影响 研究了不同浓度SO_2对大鼠胸主动脉平滑肌、内皮细胞钙离子通道的作用机制。采用剥离内膜块的组织贴块法分离大鼠主动脉EC及SMC,分别以5、15、25、50、100、500、1000、2000μmol·L~(-1) SO_2水溶液处理大鼠胸主动脉平滑肌细胞,以200、1000、2000μmol·L~(-1) SO_2水溶液处理大鼠胸主动脉内皮细胞,用激光共聚焦显微镜扫描测定细胞中的自由钙离子浓度。结果显示:①累计加入浓度为5、15、25、50、100、500、1000、2000μmol·L~(-1)SO_2水溶液后,与对照组相比,平滑肌细胞内的钙离子荧光强度分别下降到98.58%,90.10%,85.09%,79.56%,73.13%,70.21%,11.35%和8.72%。用NE(1μmol·L~(-1))预处理血管平滑肌细胞后,显著削弱了高浓度SO_2(1000μmol·L~(-1))降低[Ca~(2+)]_i的作用,但却没有削弱低浓度SO_2(10、50μmol·L~(-1))降低[Ca~(2+)]_i的作用。 ②累计加入浓度为200、1000、2000μmol·L~(-1)SO_2水溶液后,与对照组相比,内皮细胞内的钙离子荧光强度分别下降到90.90%,68.97%,7.06%。
[Abstract]:The role of lactic acid as an intermediate of glucose metabolism in the peripheral nervous system, which connects the central nervous system with all the organs of the body, including the brain and spinal nerves, has been extensively studied. The accumulation of lactic acid is considered to be one of the mechanisms of fatigue. In addition, some recent studies have found that ammonia accumulation in exercise is closely related to fatigue. Especially in some cities or regions where the industry is more developed, the population is more dense or the high sulfur coal is burned, the SO_2 exceeds the standard very seriously. Therefore, the study of the effect of so _ 2 on human health has been paid more and more attention by the society. In order to investigate the mechanism of antihypertensive effect of SO_2 on rats and the mechanism of vasodilation induced by SO-2 from the point of view of signal transduction, the effects of SO_2 on cultured smooth muscle cells and endothelial cells were measured in Wistar rats. The effects of lactic acid and ammonia on thoracic aortic vasodilation in Wistar rats were studied. 1. Effects of lactic acid and ammonia on vasodilation and their combined effects with SO_2. The effects of lactic acid and ammonia on vasodilation and their combined effects with other drugs were studied. Medlab-U / U / 4C501 biological signal acquisition and processing system was prepared to measure the tension of vascular rings in rats with different concentrations of lactic acid. At the same time, the tension of vascular ring decreased after adding different concentrations of lactic acid. But the diastolic ratio was smaller than that of lactic acid or other drugs alone, and the relaxation of the aorta rings was concentration-dependent after the addition of 0.3-20mmol 路L ~ (-1) ammonia), but the relaxation of the aorta was not affected by the low concentration of ammonia (0.3-1.0 mmol 路L ~ (-1) ~ (-1)), but there was no significant effect on the vasodilation of the aorta in rats after the addition of 0.3-20 mmol 路L ~ (-1) of ammonia. However, the effect of high concentration of ammonia (2.0-20 mmol 路L ~ (-1)) is obvious. The effect of so _ 2 on the concentration of Ca~(2 produced by vascular cells. The effects of different concentrations of SO_2 on rat thoracic aortic smooth muscle were studied. The mechanism of calcium channel in endothelial cells. Rat aortic EC and SMC were separated by tissue attachment method of stripping intima. The smooth muscle cells of rat thoracic aorta were treated with SO_2 aqueous solution of 5 ~ 152550 ~ 500 ~ 500 ~ 1000 ~ 1000 渭 mol 路L ~ (-1) SO_2, respectively. The endothelium of rat thoracic aorta was treated with 2 000 渭 mol 路L ~ (-1) mol 路L ~ (-1) so _ 2 solution, and the free calcium ion concentration was measured by laser confocal microscope scanning. The results showed that the concentration of free Ca ~ (2 +) was measured by laser confocal microscope. Pretreatment of vascular smooth muscle cells with NE(1 渭 mol 路L ~ (-1) significantly weakened the effect of high concentration SO_2(1000 渭 mol 路L ~ (-1) on [Ca~(2] _ I, but did not weaken the effect of low concentration so _ 21050 渭 mol 路L ~ (-1) on [Ca~(2]. (2) the calcium fluorescence intensity in endothelial cells decreased to 68.97% and 7.06%, respectively, compared with the control group, after the accumulative concentration of 200g / 1000 ~ 2000 渭 mol 路L ~ (-1) C _ (-1) O _ (2) was added.
