人胚胎三叉神经节细胞形态及局部定位

【摘要】: 目的:观察人胚胎三叉神经节的形态结构及研究人胚胎发出眼神经、上颌神经及下颌神经的神经元在三叉神经节内的定位。方法:收集胎龄为20周以上的胎儿9例,灌注后进行分组:形态学观察组4例,胎龄为24-28周,左侧三叉神经节进行苏木素-伊红染色,右侧进行醋酸双氧铀-柠檬酸铅染色;其余5例胎龄为20-26周标本为DiI染色晶体植入组,3例左侧眼神经植入、右侧上颌神经植入,1例双侧下颌神经植入,1例左侧下颌神经、右侧三叉神经根植入,最后通过光镜、电镜及荧光显微镜等方法进行形态学观察,拍照后在Adobe Photoshop(7.0,中文版)处理,分析结果。结果:①光镜下节细胞的胞质内可见粗大的尼氏颗粒,细胞核近圆形,呈空泡状,核仁基本居中;②电镜下节细胞的胞质内细胞器丰富,可见大量线粒体、发育较好的高尔基复合体、粗面内质网及游离核糖体,核膜凹凸不平,核孔多;③荧光显微镜下三叉神经节内可见到DiI标记的节细胞和神经纤维,节细胞的胞体呈圆形或椭圆形,大小不一,可见连于胞体的突起,在神经节内还可见到双极神经元;④人胚胎三叉神经节是沿内外方向分布的细胞团,发出眼神经的神经元位于神经节的前内侧,下颌神经的神经元位于神经节的后外侧,上颌神经的神经元位于两者之间。结论:胎龄24-28周人胚胎三叉神经元显示为成熟细胞特征;人胚胎发出眼神经、上颌神经及下颌神经的神经元在三叉神经节内以从内向外的局部定位方式排列。
[Abstract]:Objective: to observe the morphological structure of the trigeminal ganglion of human embryo and to study the localization of the neurons of the human embryonic emitting ophthalmic nerve, maxillary nerve and mandibular nerve in the trigeminal ganglion. Methods: nine fetuses over 20 weeks of gestational age were collected. After perfusion, there were 4 cases in the morphological observation group with gestational age of 24-28 weeks. The left trigeminal ganglion was stained with hematoxylin eosin, and the right side was stained with dioxy-uranium-citrate lead acetate. The other 5 cases of 20-26 weeks of gestational age were implanted with DiI staining lens. The left eye nerve was implanted in 3 cases, the bilateral mandibular nerve was implanted in 1 case and the right trigeminal nerve root was implanted in the left mandibular nerve in 1 case, and the root of the right trigeminal nerve was implanted with light microscope. Electron microscope and fluorescence microscope were used to observe the morphology of the ganglion cells. The results were analyzed by Adobe Photoshop 7.0. results under the light microscope of 1: 1, the cytoplasm of the ganglion cells could be seen in the thick Nissl granules, the nucleus was nearly round, and the cells were vacuolated. The cytoplasmic organelles of the nucleus were abundant in the inferior ganglion cells under electron microscope. A large number of mitochondria, well developed Golgi complex, rough endoplasmic reticulum and free ribosome were observed, and the ribosome was uneven. DiI labeled ganglion cells and nerve fibers could be seen in the trigeminal ganglion under the fluorescence microscope of the nuclear foramen. The ganglion cells were round or oval in size and different in size, and the processes attached to the cell bodies could be seen. In the ganglion, the trigeminal ganglion of 4 human embryos is a cell cluster distributed in the inner and outer directions. The neurons of the ophthalmic nerve are located on the anterior medial side of the ganglion, and the neurons of the mandibular nerve are located on the lateral side of the ganglion. Conclusion: the trigeminal neurons of human embryo at 24-28 weeks of gestational age show the characteristics of mature cells. The neurons of the maxillary nerve and the mandibular nerve are arranged in the trigeminal ganglion in an inward and outward local location.
