同种异体大鼠气道上皮种植模型的建立及意义

【摘要】: 目的通过同种异体大鼠气管的异位移植,建立气道上皮再生的模型。方法实验组(n=20)取Wistar大鼠气管经胰蛋白酶冷消化制成去上皮化气管作为供体,灌入含分化成熟的气管上皮细胞株等混合液,植入到受体裸大鼠背部皮下。对照组(n=20)供体制作方法同实验组,灌入等渗的生理盐水,植入到受体裸大鼠背部皮下。于移植后30天获取移植气管标本,制作病理切片及免疫组化。结果30天后获取的实验组移植气管标本病理切片HE染色见气管上皮细胞生长,免疫组织化学结果角蛋白抗体阳性。而30天后获取的对照组移植气管标本病理切片HE染色未见气管上皮细胞生长,免疫组织化学结果角蛋白抗体阴性。结论同种异体大鼠气管上皮细胞种植模型建立的成功,对肺移植或气管移植术后闭塞性细支气管炎的临床研究和治疗具有重要意义。
[Abstract]:Objective to establish a model of airway epithelium regeneration through allogeneic trachea transplantation in rats. Methods the trachea of Wistar rats was cold digested by trypsin and used as donor. A mixture of differentiated and mature tracheal epithelial cells was injected into the dorsal subcutaneous of the recipient nude rats. The donor of the control group was made in the same way as the experimental group, and the isosmotic saline was injected into the donor. The transplanted trachea specimens were obtained 30 days after transplantation, and pathological sections and immunohistochemistry were made. Results HE staining showed the growth of tracheal epithelial cells in the experimental group after 30 days. Immunohistochemical results showed that keratin antibody was positive, but 30 days later, there was no growth of tracheal epithelial cells in the pathological sections of trachea grafts in the control group. The results of immunohistochemistry showed that keratin antibody was negative. Conclusion the successful establishment of rat tracheal epithelial cell implantation model is of great significance for the clinical study and treatment of bronchiolitis obliterans after lung transplantation or trachea transplantation.
