老年肝郁失眠大鼠模型的建立及舒郁安神方作用的研究

【摘要】: 目的:建立老年肝郁失眠大鼠模型,探讨舒郁安神方治疗老年肝郁失眠的作用机制 方法:采用腹腔注射D-半乳糖致亚急性衰老,进行形态、体重观察,并测定脑组织SOD、MDA含量;夹尾刺激致肝郁,并进行形态观察以及血浆6-keto-PGF_(1a)、TXB_2的检测;改良多平台水环境法致失眠,并进行形态观察和录像监控,从而建立老年肝郁失眠大鼠模型。通过延长戊巴比妥钠睡眠时间实验和戊巴比妥钠阈下催眠剂量实验,以舒乐安定为对比,研究舒郁安神方的促眠作用。Morris水迷宫中的定位航行和空间探索实验用于测定舒郁安神方对老年肝郁失眠模型大鼠空间学习记忆能力的影响。舒郁安神方对老年肝郁失眠模型大鼠脑组织Glu及GABA含量的影响,则采用高效液相法分别测定额叶皮质及下丘脑部位Glu、GABA的含量,并与空白对照组、证候模型组及舒乐安定组作比较。 结果:衰老造模后,老年组、模型组大鼠体重增长缓慢,六周后体重与空白组对比,差异有显著性(P<0.01);脑组织SOD、MDA含量与空白组对比,差异有显著性(P<0.01),而老年组与模型组之间差别无统计学意义。肝郁造模后,肝郁组、模型组血浆TXB_2、6-keto-PGF_(1a)含量以及6-keto-PGF_(1a)/TXB_2比值与空白组对比,差异有显著性(P<0.01),而肝郁组与模型组之间差别无统计学意义。睡眠剥夺实验可观察到模型组与失眠组大鼠的总睡眠量明显减少,而空白组无此变化。促眠实验结果可见,与空白对照组比较,舒郁安神方组可协同戊巴比妥纳延长小鼠睡眠时间(P<0.05)、提高小鼠戊巴比妥钠阈下剂量时的入睡率(P<0.01);阳性对照药安定的作用较舒郁安神方的作用更加显著,均有显著差异(P<0.01)。大鼠定位航行实验中,模型组与安定组第3天、第4天的逃避潜伏期均较对照组和舒郁组所用的时间长,其两天平均成绩也明显低于对照组和舒郁组,差异均具有显著性(P<0.01),安定组与舒郁组之间则有显著性差异(P<0.01)。大鼠在空间探索实验中,模型组、安定组与对照组、舒郁组相比,20%边缘区域所占比例明显增多,40%边缘区域及中心区域所占比例明显减少,差异均具有显著性(P<0.01),而模型组与安定组之间的差异及对照组和舒郁组之间的差异则无显著性(P>0.05)。模型组大鼠额叶皮质及下丘脑部位Glu的含量与空白对照组相比,均显著升高(P<0.01);安定组大鼠各脑区Glu含量与模型组相比,均明显下降(P<0.01),舒郁组与空白对照组相比,各脑区Glu含量显著升高(P<0.01);与安定组对比,舒郁组在额叶皮质部位的含量呈现升高,差异有统计学意义(P<0.05);而在下丘脑部位的含量则明显升高(P<0.01)。与空白对照组相比,模型组、安定组大鼠额叶皮质及下丘脑部位GABA的含量显著升高(P<0.01);舒郁组与空白对照组相比,含量有所升高,差异有统计学意义(P<0.05),与模型组对比,舒郁组在脑区各部位的含量均呈现显著下降(P<0.01);舒郁组与安定组的各脑区含量比较,额叶皮质部位含量表现为下降(P<0.05),下丘脑部位则下降明显(P<0.01)。 结论:老年肝郁失眠大鼠模型的建立是较为成功的。舒郁安神方具有较好的促眠作用,对老年肝郁失眠大鼠模型的学习记忆能力具有一定的保护作用,而调节GABA、Glu两种神经递质的含量比值则可能是其治疗老年肝郁失眠的神经生物学作用机制之一。
[Abstract]:Objective: to establish a rat model of senile liver depression and insomnia, and to explore the mechanism of Shu Yu Anshen prescription in the treatment of senile liver depression and insomnia.
Methods: intraperitoneal injection of D- galactose induced aging, morphological observation, weight, and determination of brain tissue SOD, MDA content; tail pinch induced liver, and to observe the morphology and plasma 6-keto-PGF_ (1a), TXB_2 detection; modified multiple platform method caused by insomnia, and to observe the morphology and video monitoring, so as to establish a rat model of insomnia in elderly depression. By prolonging the sleeping time induced by pentobarbital sodium experiment and pentobarbital sodium subthreshold hypnotic dose experiment with estazolam for comparison, research and promotion of Shuyu positioning navigation and space effect of.Morris water maze sleep experiment for determination of Shuyu Anshen recipe learning the memory ability of aged rats. Liver depression and insomnia effect of Shuyu Anshen Decoction on Glu and GABA in the brain tissue of Elderly Rats Liver Depression Insomnia model, using HPLC and cortex were measured. The hypothalamus Glu, GABA content, and with the control group, syndrome model group and the diazepam group for comparison.
Results: the aging model, elderly group, body weight of rats in model group increased slowly, weight compared with the control group after six weeks, there was significant difference (P < 0.01); brain tissue SOD, MDA content compared with the control group, there was significant difference (P < 0.01), but no significant difference in the elderly between the group and model group. After the model of liver qi stagnation syndrome group, model group, plasma TXB_2,6-keto-PGF_ content (1a) and 6-keto-PGF_ (1a) /TXB_2 ratio compared with the control group, there was significant difference (P < 0.01), but there is no significant difference between depression group and model group. Sleep deprivation can be observed the total amount of sleep and insomnia model group rats were significantly decreased, while the control group no significant changes. Hypnogenetic experimental results show that compared with the control group, and group of Shu Yu can cooperate with sodium pentobarbital sleeping time in mice (P < 0.05), improve the pentobarbital sodium in small doses sleep 鐜,
