泡球蚴感染纯系小鼠诱导免疫耐受的实验研究

【摘要】: 目的:建立C57BL/6小鼠对BALB/c小鼠的尾部皮肤移植模型,观察健康小鼠及感染泡球蚴1月及3月移植皮片存活情况,探讨感染泡球蚴的小鼠皮肤移植后,其免疫系统的变化对移植物的影响。方法:采用小鼠尾部皮肤移植建立小鼠皮肤移植模型。实验选用64只健康的C57BL/6雄性小鼠,80只健康BALB/c雄性小鼠,随机选取其中48只接种包虫。实验分为4组,每组各16对:A组.空白对照组(非同系移植不予以任何干预):健康C57BL/6小鼠→健康BALB/c小鼠; B组.实验组1:健康C57BL/6小鼠→BALB/c小鼠(感染泡球蚴后1个月后); C组.药物对照组[非同系移植加药物干预:新型免疫抑制剂西罗莫司(雷铂霉素)]:健康C57BL/6小鼠→健康BALB/c小鼠,皮肤移植术后五天每日给予西罗莫司1.5mg/kg.d干预;D组.实验组2:健康C57BL/6小鼠→BALB/c小鼠(感染泡球蚴后3个月后)。术后观察BALB/c小鼠尾部移植皮肤生长情况,有无苍白,干瘪,结痂,毛发脱落等情况,出现上述情况视为移植皮片死亡,若出现包扎脱落等情况视为技术失败,技术失败小鼠不纳入实验中并及时补足小鼠只数。每组留8只用于观察BALB/c小鼠术后尾部移植皮肤生存期,每组于术后3、5、7、9天四个时间点各取两只小鼠尾部移植皮片,用于观察小鼠尾部移植皮肤病理变化情况。结果:A组、B组、C组、D组移植皮肤存活时间平均分为6.0±0.76、7.5±0.93、13.25±1.04、11.5±0.93天。C、D组移植皮肤存活时间较A、B组延长,差异有统计学意义(p0.01),组织病理学检查:A、B组移植皮片均在术后一周左右发生明显的逐渐加重的急性免疫排斥反应, C、D组移植皮片的免疫排斥反应较A、B组轻。结论:小鼠感染泡状棘球蚴后有利于同种异体移植皮片的存活,可以延长移植皮片的生存时间。该模型为研究感染泡球的免疫实验研究提供了良好的实验平台,为进一步研究泡球蚴感染后机体免疫状态的变化提供了基础。
[Abstract]:Objective: to establish the tail skin transplantation model of BALB/c mice in C57BL/6 mice and to observe the survival rate of transplanted skin grafts in healthy mice and infected alveolar echinococci for one month and three months, and to explore the skin transplantation of mice infected with alveolar echinococci.The effects of changes in its immune system on grafts.Methods: mouse skin transplantation model was established by tail skin transplantation.Sixty-four healthy C57BL/6 male mice and 80 healthy BALB/c male mice were selected and 48 of them were randomly inoculated with hydatid worms.The experiment was divided into 4 groups, 16 pairs in each group.Blank control group (non-homologous transplantation without any intervention: healthy C57BL/6 mice healthy BALB/c mice; group B).Experimental group 1: healthy C57BL/6 mice BALB/c mice (1 month after infection with alveolar hydatid); C group.Drug control group [non-homologous transplantation plus drug intervention: a new immunosuppressant sirolimus (rifamycin)]: healthy C57BL/6 mice were treated with sirolimus 1.5mg/kg.d for 5 days after skin transplantation.Group 2: healthy C57BL/6 mice BALB/c mice (3 months after infection with alveolar hydatid).After operation, the skin growth of BALB/c mice was observed, whether the skin was pale, dry, scabbed, hair shed and so on. The above condition was regarded as the death of skin grafts, and if the cases of bandaging and shedding were regarded as technical failure,The technique failed mice were not included in the experiment and the number of mice was replenished in time.Eight skin grafts were used in each group to observe the survival time of BALB/c mice after operation. Two mice skin grafts were taken from each group at four time points of 79-day after operation to observe the pathological changes of the skin grafted from the tail of the mice.Results the average survival time of skin graft in group C and D was 6.0 卤0.76 卤0.93 days. The survival time of skin grafts in group C was longer than that in group A (13.25 卤1.04U 11.5 卤0.93 days).The histopathological examination showed that the acute immune rejection of grafts in group 1 was significantly worse than that in group Agna B, and that in group C D was less than that in group Agna B (P < 0 01), and that in group C was significantly worse than that in group A (P < 0 01).Conclusion: infection with Echinococcus alveolaris in mice is beneficial to the survival of allograft skin grafts and can prolong the survival time of skin grafts.The model provides a good experimental platform for the study of immunological study of infected alveolar spheres, and provides a basis for further study on the changes of immune state after infection of alveolar echinococci.
