强迫游泳后孕鼠中枢ER_α的表达

【摘要】: 目的 观察孕鼠在强迫游泳实验(FST)中的不动时间,了解孕鼠是否具有更明显的行为绝望表现(即停止挣扎、不动时间延长);同时观察反复强迫游泳后孕鼠的行为绝望程度(不动时间长短)是否加重; 半定量测定强迫游泳后小鼠大脑皮质雌激素α受体(ER_α)的表达水平,比较孕鼠和非孕鼠之间的差异; 简单分析孕鼠在FST中的不动时间与中枢ER_α表达水平之间的数量关系;初步探讨小鼠FST的行为绝望程度(不动时间长短)与中枢ER_α表达水平之间是否直接相关。 材料及方法 同批次雌昆明鼠,来源于中国医科大学动物实验中心。随机分为受孕组(合笼受孕)和对照组,共获得31只孕鼠和27只对照鼠;其中有3只孕鼠早产,未进行第6次FST和免疫组化检测。 孕鼠受精后12天,开始进行FST,随机抽取非孕鼠作为对照;一共6次FST,记录第2次和第6次的不动时间。 完成6次FST后,取大脑组织,共有28只孕鼠和13只对照鼠标本;以海马为中心纵切,石蜡包埋,切片行免疫组化链霉菌抗生物素-过氧化物酶(SP)法半定量检测大脑皮质ER_α。 统计学分析采用SPSS13.0软件;计量资料不满足正态分布和方差齐性,采取Mann-Whitney U检验;满足总体正态和样本方差齐性,采取两独立样本t检验;两变量间关系采用Pearson直线相关分析。 结果 孕鼠在第2次和第6次FST中不动时间均长于非孕鼠,P分别为0.011和0.016,差异有统计学意义;孕鼠第6次FST比第2次FST不动时间长,P为0.016,差异有统计学意义。 强迫游泳后孕鼠和对照鼠大脑皮质均有ER_α表达,但孕鼠中枢ER_α组化评分高于非孕鼠,P为0.008,差异有统计学意义。 比较第6次FST不动时间t>180秒的孕鼠和t<180秒的孕鼠之间大脑皮质ER_α组化评分差异,P为0.128,无统计学意义。 小鼠两次FST的不动时间分别与中枢ER_α组化评分做散点图,均无明显直线相关趋势,R值为0.041和0.082,P值0.800和0.607,回归系数均无显著意义,不能认为有直线关系。 结论 孕鼠在FST中不动时间长于非孕鼠,行为绝望状态更明显,具有抑郁倾向;经过6次强迫游泳后孕鼠在FST中不动时间延长,可考虑作为一种产前抑郁症的模型。 强迫游泳后孕鼠中枢ER_α表达水平明显高于非孕鼠,它可能与产前抑郁症的发生有关。 孕鼠在FST中不动时间的长短与中枢ER_α表达水平之间尚未发现有数量上的联系;小鼠在FST不动时间与中枢ER_α水平之间尚未发现有直线关系,暂不能认为二者直接相关。
[Abstract]:PurposeTo observe the immobility time of pregnant rats in forced swimming experiment (FSTs), to find out if pregnant rats have more obvious behavioral despair (that is, stop struggling).The time of immobility was prolonged and the degree of behavioral despair (length of immobility) of pregnant rats after repeated forced swimming was observed.The expression level of estrogen 伪 receptor ER伪 in cerebral cortex of mice after forced swimming was measured semi-quantitatively to compare the difference between pregnant and non-pregnant mice.The quantitative relationship between the immobility time of pregnant rats in FST and the expression level of central ER- 伪 was analyzed, and the relationship between the degree of behavioral despair (immobility time) of FST and the expression level of central ER- 伪 in mice was preliminarily investigated.Materials and methodsThe same batch of female Kunming mice came from the Animal experiment Center of China Medical University.31 pregnant rats and 27 control mice were randomly divided into the pregnant group and the control group, 3 of them were preterm, and the sixth FST and immunohistochemistry were not performed.After 12 days of fertilization, FSTs were performed and non-pregnant mice were randomly selected as control, and the second and sixth immobility times were recorded for 6 FSTs.After 6 times of FST, 28 pregnant rats and 13 control mice were taken from the brain tissue, the hippocampus was cut longitudinally, embedded in paraffin, and the ER伪 in the cerebral cortex was detected by immunohistochemical streptomyces biotin peroxidase (SPSP) method.The statistical analysis adopts SPSS13.0 software; the measurement data do not satisfy the normal distribution and homogeneity of variance, the Mann-Whitney U test is used; the total normal distribution and sample variance homogeneity are satisfied, and two independent samples t test are adopted; the relationship between the two variables is analyzed by Pearson linear correlation analysis.ResultThe immobility time of pregnant rats in the second and sixth FST was 0.011 and 0.016, respectively, the difference was statistically significant, and the FST of the sixth pregnant mice was 0.016 longer than that of the second FST (P = 0.016).There was ER伪 expression in the cerebral cortex of pregnant rats and control rats after forced swimming, but the ER _ 伪 histochemical score of pregnant rats was higher than that of non-pregnant rats (P = 0.008), and the difference was statistically significant.The difference of ER伪 histochemical score between pregnant rats with FST immobility time t > 180 seconds and pregnant rats with t < 180 seconds was 0.128, which had no statistical significance.The fixed time of FST in mice and the central ER- 伪 histochemical score were divided into scattered plot. There was no obvious linear correlation R value of 0.041 and 0.082% P value 0.800 and 0.607, respectively. The regression coefficient was not significant and could not be regarded as a linear relationship.ConclusionThe immobility time of pregnant mice in FST was longer than that of non-pregnant mice, the behavior of desperate mice was more obvious, and the pregnant mice had depression tendency. After 6 times of forced swimming, the pregnant rats' immobility time in FST was prolonged, so the pregnant mice could be considered as a model of prenatal depression.The level of ER伪 expression in pregnant rats after forced swimming was significantly higher than that in non-pregnant rats, which may be related to the occurrence of prenatal depression.There was no quantitative correlation between the length of immobility time in FST and the expression level of central ER伪 in pregnant mice, but there was no linear relationship between FST immobility time and central ER伪 level in mice, so it was not possible to think that the two were directly related.
