酶免疫荧光测量中的数据处理

【摘要】: 20世纪中叶以来,一系列的免疫分析技术被逐渐创立,免疫分析技术是生物医学领域中的新方法。其特点是高特异性、高灵敏度,分析费用低廉,操作简便,被广泛应用于医学和生物学研究领域。 免疫分析技术主要包括放射免疫、酶免疫、时间分辨率荧光免疫、化学发光免疫等。本文通过介绍免疫分析技术的发展和学术背景,对各种免疫分析进行调研,了解各分析技术的原理、应用及存在的优缺点。 文章介绍了酶免疫荧光分析仪的总体结构、各组成部分及其功能。介绍了酶免疫荧光分析中广泛应用的数据处理方式,主要包括多项式拟合、log-logit拟合、三次样条插值法拟合、四参数和五参数logistic拟合,并详细介绍了这几种拟合方式的数学模型、拟合算法及在编写拟合程序中所遇到的一系列问题和解决方法。在数据处理的程序设计部分,介绍酶免疫荧光分析仪的几种拟合方法程序的程序编写、数据库的程序编写及初始显示界面的程序编写,详细的给出了程序的流程图、程序代码和采用不同的拟合程序所得到的拟合结果。其中着重对四、五参数logistic拟合的程序作了稳定性研究,研究在数据处理过程中四、五参数logistic拟合的稳定性,得出在变化范围不超过10%时,程序能很好的反应数据的走向,但当数据变化范围超过30%时,五参数拟合优度显著变劣,迭代次数显著增加,得到的曲线已经不能反映测量结果了。最后对工作完成情况做了相应的总结,指出了还未完成的部分和需要改进的部分。
[Abstract]:Since the middle of the 20th century, a series of immunoassay techniques have been gradually established, and immunoassay is a new method in biomedical field.It is characterized by high specificity, high sensitivity, low cost and simple operation, so it is widely used in medical and biological research fields.Immunoassay techniques mainly include radioimmunity, enzyme immunity, time resolution fluorescence immunity, chemiluminescence immunity and so on.In this paper, the development and academic background of immunoassay are introduced, and the principle, application, advantages and disadvantages of each analysis technique are investigated.In this paper, the structure, components and functions of enzyme immunofluorescence analyzer are introduced.This paper introduces the widely used data processing methods in enzyme immunofluorescence analysis, mainly including polynomial fitting log-logit fitting, cubic spline interpolation, four-parameter and five-parameter logistic fitting, and the mathematical models of these fitting methods are introduced in detail.Fitting algorithm and a series of problems and solutions in programming fitting program.In the part of program design of data processing, the programming of several fitting methods of enzyme immunofluorescence analyzer, the programming of database and the program of initial display interface are introduced, and the flow chart of the program is given in detail.The program code and the results obtained by using different fitting programs.The stability of logistic fitting program with four and five parameters is studied, and the stability of logistic fitting with four and five parameters in the process of data processing is studied. It is concluded that the program can well reflect the trend of data when the range of variation is less than 10.But when the variation range of the data exceeds 30, the goodness of fit of the five parameters becomes worse and the iteration times increase significantly. The obtained curve can no longer reflect the measured results.Finally, the paper summarizes the work, points out the unfinished parts and the parts that need to be improved.
