流感病毒感染诱导宿主细胞凋亡的分子机理研究以及果蝇泛素化蛋白Dhr6的初步研究

【摘要】: (一)细胞凋亡是一个非常重要的细胞生理过程,它对于维持机体自身的稳定性和个体正常的生长发育以及抑制肿瘤发生等许多方面都起着重要的作用。目前研究表明,许多病毒的感染都能够诱导宿主细胞的凋亡,但其具体的机制还不清楚,包括是病毒的什么成份引起了宿主细胞的凋亡以及宿主细胞是通过什么途径走向凋亡的。 凋亡诱导因子(apoptosis-inducing factor, AIF)和线粒体内切核酸酶G(endonuclease G, Endo G)在细胞凋亡中起到了非常重要的作用,但其发挥凋亡效应的特点还没有定论,在不同的凋亡诱导因素下可能会有不同的行为特点。本文首先证明了AIF和Endo G在流感病毒感染诱导细胞凋亡过程中起到了重要的作用,重点研究了AIF和Endo G发挥的凋亡诱导效应与caspase活化之间的关系,发现二者有着不同的作用特点。本文还研究了AIF和Endo G在诱导细胞凋亡过程中与P53活化以及Hsp70等相关蛋白的关系。 (二)蛋白质的泛素化修饰是蛋白质的一种重要的翻译后水平修饰,人们对泛素-蛋白酶体途径介导的蛋白质的降解机制也已经有了较深入地了解,但近年来发现有些蛋白质(例如组蛋白)在被泛素化修饰后并不能使蛋白质降解,而是对蛋白质的功能进行调控。 Dhr6是酵母中Rad6的在果蝇中的同源蛋白,在酵母中Rad6是催化泛素化过程中起E2作用的酶,并且在DNA损伤修复中发挥着重要的作用。但对果蝇Dhr6的研究就尚未见报道,本文重点研究了果蝇Dhr6的亚细胞定位以及对DNA损伤试剂MMS的应答特点,还研究了另外一个同样处于E2位值的泛素化酶DubcX与Dhr6的行为差别,最后优化了Dhr6的RNAi实验条件。
[Abstract]:(1) apoptosis is a very important cellular physiological process, which plays an important role in maintaining the stability of the body itself, normal growth and development of the individual, and inhibiting tumorigenesis. Current studies have shown that many viral infections can induce apoptosis in host cells, but its specific mechanism is not clear. Including what components of the virus cause the apoptosis of host cells and through what way the host cells apoptosis. Apoptosis-inducing factors (AIFs) and mitochondrial endonuclease G(endonuclease G (Endo G) play a very important role in apoptosis, but the characteristics of apoptosis-inducing factor (AIFs) and mitochondrial endonuclease G(endonuclease G (Endo G) play an important role in apoptosis, but the characteristics of apoptosis-inducing factor play an important role in apoptosis. There may be different behavioral characteristics under different apoptosis-inducing factors. In this paper, we first demonstrated that AIF and Endo G play an important role in the process of apoptosis induced by influenza virus infection, and focused on the relationship between the apoptosis-inducing effect of AIF and Endo G and the activation of caspase. It is found that the two have different characteristics. We also studied the relationship between AIF and Endo G and p53 activation, Hsp70 and other related proteins in the process of inducing apoptosis. (2) the ubiquification modification of proteins is an important post-translational modification of proteins, and the mechanism of protein degradation mediated by the ubiquitin proteasome pathway has been well understood. However, in recent years, it has been found that some proteins (such as histone) can not degrade proteins after being modified by ubiquitin, but regulate the function of proteins. Dhr6 is the homologous protein of Rad6 in Drosophila and Rad6 is the enzyme that catalyzes ubiquification and plays an important role in the repair of DNA damage. However, the study of Drosophila Dhr6 has not been reported. In this paper, the subcellular localization of Drosophila Dhr6 and its response to DNA damage reagent MMS were studied, and the behavioral differences between DubcX and Dhr6, another ubiquitin enzyme at E2 level, were also studied. Finally, the RNAi experimental conditions of Dhr6 are optimized.
