广西10例人芽囊原虫基因型和同工酶谱的研究

【摘要】:目的分析广西10个人芽囊原虫(Blastocystis hominis)分离株基因型,及其乳酸脱氢酶(LDH)和酯酶(EST)的同工酶谱特征。方法从感染者粪便中分离到的10个人芽囊原虫(BhGX1~BhGX10),体外培养并提取基因组DNA。用已知的7对特异性序列标记位点(sequene tagged sites,STS)引物PCR扩增,来鉴定基因型。采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE),分别进行乳酸脱氢酶和酯酶2种同工酶染色,比较分析10个分离株的酶谱。结果 10个人芽囊原虫分离株中有8个为基因Ⅰ型;另外2个分离株(BhGX4和BhGX7)经7对引物扩增均为阴性,为未知基因型。10个人芽囊原虫分离株在LDH谱中共出现10条酶带,常见酶带为Rm37、Rm49、Rm57、Rm68和Rm92;在EST谱中共出现12条酶带,其中Rm14、Rm18、Rm23、Rm27、Rm45、Rm50和Rm77酶带较常见。各分离株之间的LDH和EST同工酶谱均存在差异。结论广西10个人芽囊原虫分离株以基因Ⅰ型为主,但各分离株之间的LDH和EST同工酶谱存在差异。
[Abstract]:Objective to analyze the isozyme patterns of lactic dehydrogenase (LDH) and esterase (EST) of 10 isolates of Blastocystis hominis from Guangxi. Methods BhGX1 (BhGX1) BhGX10 was isolated from the feces of infected individuals, and the genomic DNA was cultured and extracted in vitro. The genotypes were identified by PCR amplification using 7 pairs of known primers for sequene tagged sitesmotif. Two isozymes of lactate dehydrogenase and esterase were stained by sodium dodecyl sulfate and polyacrylamide gel electrophoresis (SDS-PAGEG). The zymograms of 10 isolates were compared and analyzed.
