两种脾虚证模型对小鼠骨骼肌形态和功能的影响

【摘要】:目的探讨脾虚证与骨骼肌收缩功能的关系。方法将36只ICR小鼠按体重配对原则随机分成3组,即对照组、劳倦过度组及大黄组,每组12只。其中劳倦过度组及大黄组应用大黄泻下法和力竭游泳加剥夺睡眠的方法制备两种脾虚证动物模型;采用m-ATPase酶组织化学方法测定3组小鼠趾长伸肌(extensor digitorum longus,EDL)和比目鱼肌(soleus,SOL)m-ATPase酶活性的变化,计算Ⅰ型和Ⅱ型肌纤维的比例;Power Lab系统测定EDL和SOL等长收缩和强直收缩的最大张力。结果劳倦过度组及大黄组小鼠的体重、体温与一般健康状况均明显下降;脾脏、胸腺脏器指数亦低于对照组;等长收缩最大张力和强直收缩最大张力明显下降;EDL和SOL的肌纤维横截面积减小,细胞排列松散;EDL中Ⅰ型肌纤维比例增加,Ⅱ型肌纤维比例降低,SOL中Ⅰ型和Ⅱ型肌纤维比例无变化。结论劳倦过度组及大黄组小鼠的EDL和SOL均出现明显萎缩,劳倦过度组SOL中Ⅰ型肌纤维萎缩更加明显。
[Abstract]:Objective to study the relationship between spleen deficiency syndrome and skeletal muscle contraction function. Methods Thirty-six ICR mice were randomly divided into 3 groups according to the principle of body weight pairing: control group, overworked group and rhubarb group, 12 mice in each group. Two kinds of spleen deficiency syndrome animal models were established in the overworked group and rhubarb group by the method of rhubarb diarrhea and exhaustive swimming plus sleep deprivation. The enzyme activities of extensor digitorum longus (EDL) and soleus muscle (Soleusus soleus) were determined by m-ATPase histochemical method.
