兔牙周炎与动脉粥样硬化结合模型的建立及分析

【摘要】:目的:建立兔牙周炎与动脉粥样硬化结合模型,并对其进行分析,探讨牙周炎与动脉粥样硬化间的联系。方法:48只雄性新西兰兔随机分为4组:对照组(A组),牙周炎组(B组),动脉粥样硬化组(C组),牙周炎+动脉粥样硬化(D组)。各组分别按照实验设计进行相应处理,检测牙周指标、血脂指标及血清中高敏C反应蛋白、基质金属蛋白酶9的含量,12周末处死动物,X线观察实验牙牙槽骨吸收情况,取升主动脉行HE及Masson染色,进行斑块面积及稳定性评估,免疫组化染色检测斑块内基质金属蛋白酶9、单核细胞趋化蛋白-1含量。结果:B、D组探诊深度和龈沟出血指数均高于A、C组(P0.01),X线见实验牙牙槽骨吸收明显,C、D组血脂指标均高于A组及B组(P0.01),病理见C、D组动脉粥样硬化形成,同时D组病变较C组更加严重,斑块更加不稳定。血清高敏C反应蛋白、基质金属蛋白酶9含量D组最高,C、B、A组依次减少。免疫组化染色见D组斑块内基质金属蛋白酶9、单核细胞趋化蛋白-1表达多于C组(P0.01),同时可见B组内膜少许表达。结论:牙周炎可以加重动脉粥样硬化的病变程度并使斑块更加不稳定,这种作用可能与牙周炎时体内更加严重的炎症状态,及斑块局部基质金属蛋白酶9、单核细胞趋化蛋白-1等细胞因子表达水平的升高有关。
[Abstract]:Aim: to establish a rabbit model of periodontitis and atherosclerosis and analyze the relationship between periodontitis and atherosclerosis. Methods Forty-eight male New Zealand rabbits were randomly divided into four groups: control group A, periodontitis group B, atherosclerosis group C, periodontitis atherosclerosis group D. The periodontal index, blood lipid index, Gao Min C-reactive protein and matrix metalloproteinase-9 in serum were detected in each group according to the experimental design. After 12 weeks, animals were killed to observe the alveolar bone resorption. The ascending aorta was stained with HE and Masson, and the plaque area and stability were evaluated. The contents of matrix metalloproteinase 9 and monocyte chemoattractant protein-1 were detected by immunohistochemical staining. Results the depth of probing and the bleeding index of gingival sulcus in group D were higher than those in group A (P 0.01). The blood lipid indexes in group C were higher than those in group A and group B, and the atherosclerosis was found in group C, and the lesion in group D was more serious than that in group C. Plaque is more unstable. Serum Gao Min C-reactive protein and matrix metalloproteinase 9 content in group D were decreased in turn. Immunohistochemical staining showed that matrix metalloproteinase 9 and monocyte chemoattractant protein-1 were more expressed in plaque of group D than those in group C (P 0.01) and a little in group B. Conclusion: periodontitis can aggravate the severity of atherosclerosis and make the plaque more unstable, which may be related to the more severe inflammation in the body during periodontitis. The expression of matrix metalloproteinase 9 and monocyte chemoattractant protein-1 were related to the increase of matrix metalloproteinase 9 and monocyte chemoattractant protein-1.
