慢性牙周炎和动脉粥样硬化动物模型的建立及TNF-α、MMP-3检测与分析

【摘要】: 目的:选取Wistar大鼠为实验对象,通过建立慢性牙周炎和动脉粥样硬化结合的动物模型并进行相关性的初步分析,探讨慢性牙周炎和动脉粥样硬化发病相关的可能机制。方法:雄性Wistar大鼠(体重200±20g)60只随机分为四组:A对照组,B慢性牙周炎组,C动脉粥样硬化组,D慢性牙周炎+动脉粥样硬化组。各组按照实验设计进行相应干预处理,至12周末处死动物,分别检查记录牙周指标,检测血清中的血脂水平,酶联免疫吸附法(ELISA)检测血清中的肿瘤坏死因子α(TNF-α)以及基质金属蛋白酶-3(MMP-3)的含量,免疫组化技术分别检测MMP-3在牙周和腹主动脉组织中的表达,并进行图像半定量分析,同时观察牙周和腹主动脉的组织病理学改变。数据进行统计学分析。结果:经过牙周检查、血脂、ELISA检测以及组织病理学观察,已成功建立了动物模型。其中D组的血脂水平、血清TNF-α及MMP-3含量明显升高,各实验组与对照组比较差异均有统计学意义(P0.05)。经过免疫组化检测发现,MMP-3在B组和D组的牙周组织中,主要在牙龈上皮呈阳性表达,并且D组水平高于B组;在实验组的腹主动脉中,主要在内皮细胞、巨噬细胞、平滑肌细胞呈阳性表达,并且D组水平高于C组,差异均有统计学意义(P0.05)。MMP-3在组织中的表达与血清中的规律相一致。MMP-3与血脂的相关性分析呈现出不相关。结论:慢性牙周炎与动脉粥样硬化发病可能具有一定的相关性,可能对动脉粥样硬化和冠心病的发生发展有重要的影响。MMP-3是独立于血脂之外的动脉粥样硬化危险因素。
[Abstract]:Objective: to investigate the possible mechanism of chronic periodontitis and atherosclerosis by establishing the animal model of chronic periodontitis and atherosclerosis in Wistar rats. Methods: male Wistar rats (body weight 200 卤20g)60) were randomly divided into four groups: control group (n = 4) and chronic periodontitis group (n = 4). According to the experimental design, the animals were killed at the end of 12 weeks. The periodontal indexes were recorded and the serum lipids were measured. Serum levels of TNF- 伪 and matrix metalloproteinase-3 (MMP-3) were detected by Elisa, and the expression of MMP-3 in periodontal and abdominal aorta was detected by immunohistochemistry. The histopathological changes of periodontal and abdominal aorta were observed. The data were analyzed statistically. Results: the animal model was successfully established by periodontal examination, Elisa and histopathological observation. The levels of blood lipid, TNF- 伪 and MMP-3 in group D were significantly higher than those in control group (P 0.05). The expression of MMP-3 in the periodontal tissues of group B and group D was mainly positive in gingival epithelium, and the level of MMP-3 in group D was higher than that in group B. in the abdominal aorta of experimental group, the expression of MMP-3 was mainly in endothelial cells and macrophages. The expression of smooth muscle cells was positive and the level of group D was higher than that of group C. the difference was statistically significant. The expression of P0.05U. MMP-3 was consistent with the rule of serum. There was no correlation between MMP-3 and serum lipids. Conclusion: chronic periodontitis may be associated with the pathogenesis of atherosclerosis, which may have an important effect on the occurrence and development of atherosclerosis and coronary heart disease. MMP-3 is a risk factor of atherosclerosis independent of blood lipids.
