胶原材料与生长因子结合促进血管再生

【摘要】: 在组织再生与创伤修复过程中,血管再生是一个非常重要的方面,它不仅可以加速组织再生,同时也是维持肉芽组织和新生组织的必要基础。为了促进血管再生的进程,许多血管诱导生长因子与功能生物材料在组织再生及创伤修复中得到广泛应用。血小板源生长因子(Platelet-derived growth factor,PDGF)与碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)是血管诱导因子,能够诱导内皮细胞、血管平滑肌细胞、单核细胞、粒细胞以及纤维原细胞的增殖及迁移,在组织再生与创伤修复中具有重要的作用。但是,由于其在体内易扩散,以及缺少有效的传输系统限制了它们的临床应用。骨胶原(Demineralized bonematrix,DBM),即脱钙骨基质,是目前骨组织工程中最为广泛使用的支架,其主要成分为Ⅰ型胶原。脱钙骨基质除具有成骨诱导和成骨传导作用之外,其天然孔隙及连通、合适孔隙率有利于种子细胞黏附、迁移及血管的长入。 因此,为了促进组织再生与创伤修复中血管再生,我们将肝素(heparin)交联到骨胶原(HC-DBM)上,分别通过肝素特异的结合PDGF和bFGF,从而构成PDGF和bFGF的特异的靶向传输系统。在体外的实验中,肝素能够提高结合到骨胶原上生长因子的量;在体外活性实验中,结合到胶原上的生长因子能够促进成纤维细胞的快速增殖;在体内皮下包埋实验中,结合到HC-DBM上的生长因子能够促进细胞迁移到材料中;而且,组织学分析表明,HC-DBM能够促进血管再生,并能够维持血管的稳定。总之,HC-DBM能够阻止生长因子的扩散,延长其生物活性,并且能够促进材料的细胞化与血管化。
[Abstract]:Vascular regeneration is a very important aspect in the process of tissue regeneration and wound repair. It not only accelerates tissue regeneration, but also is the necessary basis for maintaining granulation tissue and new tissue. In order to promote the process of vascular regeneration, many vascular inducible growth factors and functional biomaterials have been widely used in tissue regeneration and wound repair. Platelet-derived growth factor (PDGFs) and basic fibroblast growth factor (basic fibroblast growth factor-bFGFs) are vascular inducible factors, which can induce proliferation and migration of endothelial cells, vascular smooth muscle cells, monocytes, granulocytes and fibroblasts. It plays an important role in tissue regeneration and wound repair. However, their clinical application is limited by their easy diffusion in vivo and the lack of effective transmission systems.
