长期压力超负荷大鼠心脏去甲肾上腺素转运蛋白、神经生长因子及其受体TrKA表达的变化

【摘要】: 目的:检测长期心脏压力超负荷(pressure overload,POL )大鼠心脏交感神经去甲肾上腺素转运蛋白(norepinephrine transporter, NET)表达与心肌神经生长因子(nerve growth factor, NGF)表达的关系,探讨NGF对NET的影响。 方法: 40只雄性Wistar大鼠随机分为:正常组(n=10)和POL组(n=30)二组,采用腹主动脉缩窄术建立大鼠心脏POL模型。手术8周后,用超声心电图测定大鼠心功能指标(IVSd、IVSs、IVIDd、IVIDs、LVPWd、EDV、ESV、EF、SV、FS%),计算左心室重量指数(LVMI),观察心脏结构变化,评价心脏功能。取左心室心肌组织切片,用免疫荧光方法观察左心室心肌NGF蛋白表达变化。取心脏交感神经节,即双侧颈中-星状神经节复合体(MC-SG complex,MC-SGC),及左室心肌组织,用实时荧光定量PCR检测MC-SGC中NET mRNA表达水平,及左室GAP43(growth associated factor, GAP43)mRNA、TrkA mRNA表达水平。 结果:与正常组比较,超声心动图检测到压力负荷组大鼠IVSs、LVPWd、IVIDs显著增厚;LVMI升高33.23%(p0.05);心功能(EF、FS%)增强(p0.05),表明POL大鼠处于心力衰竭代偿期。免疫荧光检测结果显示POL组心肌NGF蛋白表达增加(p0.05)。与正常对照组大鼠比较,Real Time PCR检测POL大鼠MC-SGC中NET mRNA表达水平未见显著变化;心肌GAP43 mRNA表达水平亦未见显著变化,但TrKA mRNA表达水平显著下降(p0.05)。 结论:本实验结果表明,代偿期长期压力超负荷大鼠心肌NGF蛋白表达增加,但心肌GAP43mRNA、心脏交感神经NET mRNA表达水平无明显变化,TrKA mRNA表达降低。提示(1)NET mRNA转录后异常可能是导致POL后NET功能异常的基础;(2)心肌NGF表达增高未增加神经生长,TrKA mRNA低表达水平可能是影响NGF发挥作用的重要因素。
[Abstract]:Aim: to investigate the relationship between the expression of cardiac norepinephrine transporter (NET) and the expression of nerve growth factor (NGF) nerve (NGF) nerve growth factor, NGF) in long-term cardiac pressure overload (NGF) rats, and to explore the effect of NGF on NET. Methods: forty male Wistar rats were randomly divided into two groups: normal group (n = 10) and POL group (n = 30). The rat heart POL model was established by abdominal aortic coarctation. After 8 weeks of operation, echocardiographic electrocardiogram was used to measure the cardiac function of rats. The left ventricular mass index (LVMIA) was calculated and the changes of cardiac structure and cardiac function were evaluated. The changes of NGF protein expression in left ventricular myocardium were observed by immunofluorescence. The cardiac sympathetic ganglion, MC-SG complex MC-SG complex, and left ventricular myocardial tissue were collected to detect the expression of NET mRNA in MC-SGC and the expression of GAP43(growth associated factor, GAP43mRNA-TrkA mRNA in left ventricle by real-time fluorescence quantitative PCR. Results: compared with the normal group, echocardiography showed that the IVSsl LVPWdN IVIDs were significantly thickened and LVMI was increased by 33.233p0.05, and the cardiac function was enhanced by FSI (P 0.05), indicating that the POL rats were in the compensatory phase of heart failure. The results of immunofluorescence assay showed that the expression of NGF protein increased in POL group. Compared with the control group, the expression of NET mRNA in MC-SGC of POL rats was not significantly changed by Real Time PCR, and the expression of GAP43 mRNA in myocardium was not changed significantly, but the expression of TrKA mRNA decreased significantly (p0.05). Conclusion: the results of this study indicate that the expression of NGF protein in myocardium of compensatory long-term pressure overload rats is increased, but the expression level of NET mRNA in cardiac sympathetic nerve does not change significantly. However, the expression of Trka mRNA in cardiac sympathetic nerve does not change significantly. It is suggested that the posttranscriptional abnormality of net mRNA may be the basis of NET dysfunction after POL. The increase of NGF expression in myocardium does not increase the expression of Trka mRNA, which may be an important factor affecting the role of NGF.
