脑缺血再灌流后大鼠大脑皮质微血管密度的体视学研究

【摘要】: 为了研究脑缺血再灌流后大鼠大脑皮质微血管的变化。本实验采用42只SD大鼠,分为对照组、脑缺血15min再灌流6h、12h、24h组与脑缺血2h再灌流6h、12h、24h组。用结扎左颈总动脉,夹闭右颈总动脉,再恢复右颈总动脉血流的方法建立大鼠脑缺血再灌流模型。以碱性磷酸酶法显示微血管并应用体视学方法测算大鼠海马、齿状回及顶叶皮质微血管长度密度(Lv)、表面积密度(Sv)、体积密度(Vv)的变化。结果如下: 1、各组大鼠海马各区、齿状回及顶叶皮质区微血管通过碱性磷酸酶法显示呈点状、线状或短树枝状。 2、脑缺血15min再灌流后6h、12h和24h组同对照组比较:大鼠大脑皮质微血管的Lv、Sv、Vv随着存活时间的不同而发生显著变化,尤其在再灌流后6h、12h。与对照组相比,Sv在6h、12h、24h减小(P<0.01),Lv在6h极显著下降(P<0.01),12h极显著上升(P<0.01)。Vv在6h极显著上升(P<0.01),12h显著上升(P<0.05)。24h组Lv、Vv同对照组相比无显著性变化(P>0.05)。与6h相比,12h组齿状回及顶叶皮质区微血管Vv具极显著性差异(P<0.01),24h组大脑皮质微血管Vv具极显著性差异(P<0.01)。 3、脑缺血2h再灌流后6h、12h、24h组同对照组比较:6h至24h大鼠大脑皮质微血管Lv、Sv显著低于对照组(P<0.01)。Vv在6h极显著上升(P<0.01),至24h持续高于对照组(P<0.01)。 4、脑缺血15min与脑缺血2h在同一时相点比较:6h、12h、24h大鼠大脑海马、齿状回及顶叶皮质微血管Lv、Sv、Vv均有显著性差异(P<0.01)。 5、各组中大鼠左右两侧同区比较微血管密度均不存在显著性差异(P>0.05)。各组中顶叶皮质区微血管密度高于海马各区及齿状回(P<0.01)。
[Abstract]:To study the changes of cerebral cortex microvessels after cerebral ischemia reperfusion in rats. 42 Sprague-Dawley rats were divided into two groups: the control group, the brain ischemia 15min reperfusion group (6h) and the cerebral ischemia group (12h), the control group (6h) and the control group (12h). The rat model of cerebral ischemia reperfusion was established by ligating the left common carotid artery, clipping the right common carotid artery and restoring the blood flow of the right common carotid artery. The changes of microvascular length density, surface area density and volume density in hippocampus, dentate gyrus and parietal cortex of rats were measured by alkaline phosphatase method. The results are as follows: 1. The microvessels in hippocampus, dentate gyrus and parietal cortex of each group were shown to be punctate, linear or short dendritic by alkaline phosphatase method. 2. Compared with the control group at 12 h and 24 h after 15min reperfusion, the VV v of rat cortical microvessels changed significantly with the survival time, especially at 6 h and 12 h after reperfusion. Compared with the control group, Sv decreased significantly at 6h and 12h and decreased significantly (P < 0.01) at 6h. There was no significant difference in LvVV between the control group and the control group (P < 0.01). There was no significant difference in the LvVV between the two groups (P < 0.01) and the control group (P > 0.05). There was significant difference in VV between 12 h group and 12 h group compared with 6 h group (P < 0.01). There was significant difference in VV value between 12 h group and 24 h group (P < 0.01). 3. Compared with the control group, the Sv of the cerebral cortex microvessel in the control group was significantly lower than that in the control group (P < 0.01) at 6 h to 24 h after cerebral ischemia and reperfusion (P < 0.01), and remained higher than that in the control group (P < 0.01) at 24 h. 4. There was a significant difference between 15min and VV in hippocampus, dentate gyrus and parietal cortex of rats at the same time (P < 0.01). 5. There was no significant difference in microvessel density (P > 0.05). The microvessel density in middle parietal cortex was higher than that in hippocampus and dentate gyrus (P < 0.01).
