隔药饼灸对功能性消化不良肝郁脾虚模型大鼠海马盐皮质激素受体、糖皮质激素受体、5-羟色胺1A受体表达的影响

【摘要】：目的:观察隔药饼灸对功能性消化不良(functional dyspepsia,FD)肝郁脾虚模型大鼠海马盐皮质激素受体(mineralocorticoid receptor,MR)、糖皮质激素受体(glucocorticoid receptor,GR)、5-羟色胺1A受体(5-hydroxytryptamine receptor 1A,5-HTR1A)表达的影响.方法:将50只SD大鼠按随机数字表法随机分为空白组、模型组、隔药饼灸组、逍遥散组、多潘立酮组5组,每组10只,空白组正常喂养,其余4组采用复合病因造模法(慢性束缚应激+过度疲劳+饮食失节+夹尾+摇晃)造模,连续3 wk;再按被试因素予以隔药饼灸和逍遥散、多潘立酮灌胃,共治疗2 wk.实验结束后采用营养性半固体糊灌胃法测定大鼠胃排空率,用实时荧光定量PCR(real-time fluorescence quantitative PCR,RT-PCR)法检测大鼠海马MR、GR、5-HTR1A的表达.结果:(1)模型组大鼠胃排空率较空白组明显下降(P0.01),治疗后各治疗组大鼠胃排空率较模型组上升(均P0.01),但3组间比较差异无统计学意义(P0.05);(2)与空白组比较,模型组大鼠MR表达差异无统计意义(P0.05);而G R及5-H T R1A的表达下降且差异均有统计意义(均P0.05);与模型组比较,隔药饼灸组、逍遥散组、多潘立酮组大鼠海马MR的表达亦无统计意义(P0.05),而GR及5-HTR1A的表达上调且差异均有统计学意义(均P0.05),但3组间比较差异均无统计学意义(P0.05).结论:隔药饼灸同逍遥散及多潘立酮一样能调节海马GR、5-HTR1A的表达,从而调节边缘系统-下丘脑-垂体-肾上腺轴(limbic system-hypothalamic-pituitary-adrenal axis,LHPA/HPA轴),提高大鼠胃排空率,改善胃肠动力,最终起到治疗本病的作用.
[Abstract]:Objective: to observe the effect of herbal cake moxibustion on the expression of 5-hydroxytryptamine receptor 1A (5-hydroxytryptamine receptor 1A) and glucocorticoid receptor (glucocorticoid receptor) in hippocampus of rats with liver stagnation and spleen deficiency model of functional dyspepsia. Methods: fifty Sprague-Dawley rats were randomly divided into blank group, model group, moxibustion group, Xiaoyao powder group, doperidone group, 10 rats in each group. The other four groups were made model by compound etiological method (chronic restraint stress excessive fatigue diet disjointed tail shake) for 3 wk, and then given moxibustion and Xiaoyao powder by medicine separated cake and Xiaoyao powder according to the factors of the test, and treated with domperidone for 2 wk. At the end of the experiment, the gastric emptying rate of rats was measured by semi-solid paste method, and the expression of 5-HTR1A in rat hippocampus was detected by real-time fluorescence quantitative PCR(real-time fluorescence quantitative PCR RT-PCR method. Results the gastric emptying rate in the model group was significantly lower than that in the blank group, and the gastric emptying rate in each treatment group was higher than that in the model group after treatment (all P 0.01, but there was no significant difference among the three groups) and there was no significant difference between the three groups and the blank group. There was no significant difference in Mr expression between model group and model group (P 0.05), but the expression of G R and 5 H T R1A decreased and the difference was statistically significant (all P 0 05); compared with the model group, the moxibustion group and Xiaoyao powder group were divided into two groups: moxibustion group and Xiaoyao powder group. There was no statistical significance in the expression of Mr in hippocampus of rats in domperidone group. However, the expression of gr and 5-HTR1A was up-regulated and the difference was statistically significant (P 0.05), but there was no significant difference among the three groups. Conclusion: herbal cake moxibustion, like Xiaoyao Powder and domperidone, can regulate the expression of GRN5-HTR1A in hippocampus, thus regulating limbic system-hypothalamic-pituitary-adrenal axis-LHPA / HPA axis of limbic hypothalamus-pituitary-adrenal axis, increasing gastric emptying rate and improving gastrointestinal motility in rats. Finally play a role in the treatment of this disease.
