胚胎期小鼠皮肤与羊水接触神经末梢发育规律研究

【摘要】: 研究目的 研究妊娠不同时期胚胎小鼠皮肤表面神经末梢发育规律,以及其与羊水接触的接触情况,探讨其与羊水的相互关系,分析羊水中具体物质对于皮肤表皮表面神经发育的作用。 研究方法 分别取胚胎12、13、14、15、16、17 d小胚鼠,冰冻切片,行神经特异性标记物NF免疫组织化学染色,在光镜下观察皮肤表面神经末梢的分布情况并摄片;为了进一步阐明胚胎期小鼠皮肤神经末梢是否与羊水接触,将荧光金在体注射到胚胎羊水中,切片后用荧光显微镜观察神经元被标记情况。 研究结果 胚胎期小鼠表皮表面存在有大量的神经末梢,胚胎第12d已经可以清晰看到,以胚胎第15 d最多。在胚胎的背根神经节,神经根及腹背侧灰质中均发现有明显的荧光标记。 研究结论 胚胎期小鼠皮肤神经末梢与羊水有一定的接触关系。胚胎时期小鼠皮肤神经末梢伸入羊水中,与羊水直接接触,可以感知羊水成分的变化,或者是向羊水中释放某些物质,但是羊水成分中何种物质对于周围神经生长起作用以及其起作用的具体机制现在尚不明了,需进一步探讨。
[Abstract]:Objective to study the development rule of skin surface nerve endings in different stages of pregnancy and its contact with amniotic fluid, and to explore the relationship between them and amniotic fluid. To analyze the effect of specific substances in amniotic fluid on the development of skin epidermal nerve. Methods the embryonic mice were collected from 12D embryo, frozen sections were used for NF immunohistochemical staining, the distribution of nerve endings on the skin surface was observed under light microscope, and the results were taken by radiograph. The distribution of the nerve endings on the surface of the skin was observed under light microscope, and the distribution of the nerve endings on the surface of the skin was also observed under light microscope. In order to further elucidate whether the skin nerve endings of embryonic mice are in contact with amniotic fluid, fluorescent gold was injected into embryonic amniotic fluid in vivo, and the labeling of neurons was observed by fluorescence microscope. The results showed that there were a lot of nerve endings on the surface of mouse epidermis at embryonic stage, which could be seen clearly on the 12th day of embryo, and the highest number was on the 15th day of embryo. Obvious fluorescent labeling was found in dorsal root ganglion, nerve root and ventral dorsal gray matter of embryo. Conclusion there is a certain contact relationship between skin nerve endings and amniotic fluid in embryonic mice. At embryonic stage, the skin nerve endings of mice extend into amniotic fluid and contact with amniotic fluid directly, which can sense the changes of amniotic fluid composition, or release certain substances into amniotic fluid. However, it is not clear which substances in amniotic fluid play an important role in the growth of peripheral nerve and the mechanism of its action.
