版纳微型猪近交系动物瘢痕模型的建立(实验Ⅱ)

【摘要】: [目的]以云南特有的版纳微型猪近交系为对象,建立由动物自身产生的增生性瘢痕模型,为研究增生性瘢痕的发生机制及防治措施提供较为理想的动物模型。 [方法]于10只版纳微型猪的10个背侧制造切割伤创面(左右随机):切除4cm×5cm的范围的全层皮肤及肌筋膜层,每侧2个,共计20个创面;另10个背侧制造4cm×5cmⅢ°度烧伤创面,每侧2个,共计20个创面。术后予80万单位青霉素钠注射剂肌注,每日两次,连续三日,隔离饲养。 (1)术后连续12周观察创面的自然愈合情况。 (2)对形成增生块的创面进行大体观察、组织学检测等并加入TGF-β_1及INF-γ观察其对增生块的作用。 [结果](1)切割伤创面均未见明显增生块形成,术后21天左右脱痂后创面于一周内迅速缩小,6周后创面完全上皮化,有毛发长出。 (2)烧伤创面中90%发生形态与人增生性瘢痕类似的增生块。 (3)烧伤创面增生块组织学检查见真皮中有大量成纤维细胞增殖并有少量胶原沉积,随着时间推移,胶原成份增多,真皮层内有胶原特征性的结节或漩涡状结构。 (4)局部注入TGF-β_1及INF-γ可促进或抑制增生块的发生。 [结论]版纳微型猪近交系背侧皮肤Ⅲ°烧伤后的创面经自然愈合后可发生与人增生性瘢痕类似的病理改变,该模型是研究增生性瘢痕的发生机制及评估其治疗方法的较好的动物模型之一。
