正常雌性大鼠尿道横纹括约肌肌球蛋白重链同源分子基因表达的研究

【摘要】:目的:尿道括约肌系统是哺乳类动物实现控尿的基本解剖结构,其中环绕尿道的横纹括约肌是构成尿道括约肌系统的最重要结构成分。尿道括约肌系统的控尿功能表现得复杂而精细,以满足动物在复杂多样的日常活动中对控尿的需求。研究尿道横纹括约肌纤维的类型和相对含量,可以更好地理解尿道横纹括约肌发挥复杂而精细的控尿功能的生理机制。 方法:取正常成年未生育雌性Fisher 344大鼠12只,年龄12月,麻醉后解剖取出完整尿道,提取尿道总RNA,采用逆转录-聚合酶链反应(RT-PCR)法检测尿道横纹括约肌肌球蛋白重链同源分子mRNA的类型和相对含量,从而获得尿道横纹括约肌纤维类型及其相对含量的资料。 结果:正常成年未生育雌性大鼠尿道横纹括约肌表达四种肌球蛋白重链同源分子mRNA,即MHC-Ⅰ、MHC-Ⅱa、MHC-Ⅱb和MHC-Ⅱx。其中MHC-Ⅱb的mRNA相对含量最高(38.1±0.7)%,MHC-Ⅱa次之(34.6±0.8)%,MHC-Ⅱx再次之(18.1±1.2)%,MHC-Ⅰ最低(9.2±0.4)%。 结论:正常成年未生育雌性大鼠尿道横纹括约肌表达全部4种肌球蛋白重链同源分子,显示尿道横纹括约肌含有4种类型的纤维:typeⅠ(慢收缩纤维)、typeⅡa(快收缩抗疲劳纤维)、typeⅡb(快收缩易疲劳纤维)和typeⅡx(超快收缩纤维)。尿道横纹括约肌多样化的纤维类型是其实现复杂和精细控尿功能的分子基础。
