人外泌汗腺上皮细胞原代培养的改良

【摘要】: 目的对人外泌汗腺上皮细胞原代培养的传统技术进行改良,简化实验操作,减轻劳动强度,以期用更短的时间获得更多量稳定生长的外泌汗腺上皮细胞,为体外研究汗腺和构建功能性组织工程皮肤提供理论依据和技术支持。 方法在传统的汗腺组织块培养法基础上,于分离的人外泌汗腺组织块表面加盖玻片以固定组织块,以促进汗腺组织块贴壁,减少换液等操作所致的组织块损失,并简化实验步骤。 结果改良的实验方法能显著提高汗腺组织块的贴壁率,且对汗腺上皮细胞的增殖能力无明显影响,因而在相同时间内获得的汗腺上皮细胞数量比传统方法更多。 结论与传统的组织块培养法相比,本方法的实验步骤简单,耗时更少,在相同单位时间内获得的细胞数量更多。培养的人外泌汗腺上皮细胞保持了上皮细胞的结构特点,且具有良好活力,说明了人工构建外泌汗腺的可行性。
[Abstract]:Objective to improve the traditional technique of primary culture of human exocrine sweat gland epithelial cells, simplify the experimental operation and reduce the labor intensity, so as to obtain more stable growth of exocrine sweat gland epithelial cells in a shorter time. To provide theoretical basis and technical support for the study of sweat glands and the construction of functional tissue engineering skin in vitro. Methods on the basis of the traditional culture method of sweat gland tissue block, a cover glass was added on the surface of the human exocrine sweat gland tissue block to fix the tissue mass, so as to promote the adhesion of the sweat gland tissue mass to the wall and reduce the loss of tissue mass caused by the operation of fluid exchange. The experimental steps are simplified. Results the modified method could significantly increase the adherent rate of sweat gland tissue mass, and had no obvious effect on the proliferation of sweat gland epithelial cells. Therefore, the number of sweat gland epithelial cells obtained at the same time was more than that of the traditional method. Conclusion compared with the traditional tissue mass culture method, this method has the advantages of simple experimental steps, less time consuming and more cells obtained in the same unit time. The cultured human eccrine sweat gland epithelial cells maintain the structural characteristics of epithelial cells and have good vitality, which indicates the feasibility of artificial construction of exocrine sweat glands.
