雌激素制备大鼠子宫肌瘤动物模型

【摘要】:[背景]子宫肌瘤是女性生殖系统中最常见的一种良性肿瘤,目前对于子宫肌瘤的发病机制尚不十分清楚。资料表明雌激素是肌瘤生长的主要促进因素。目前尚缺乏适宜的动物模型可供临床发病机制研究及药物筛选,建模的给药剂量及给药周期尚存在争议,值得进一步探索。 [目的]建立适合于临床发病机制研究和药物筛选的子宫肌瘤动物模型。 [方法]选取性成熟大鼠,随机分组给予200μg和300μg剂量雌激素(E2)诱导,并于8wk和10wk时处死部分动物,测定血清中雌激素(E2)和孕激素(P)的水平,观察子宫形态的变化,计算肌瘤形成率和动物死亡率,并与人子宫肌瘤进行病理组织学特征的比较,同时测定组织中雌、孕激素受体(ER、PR)的表达。确定E2诱导大鼠子宫肌瘤形成的最佳剂量和给药周期。 [结果]肌肉注射E2200μg只,2次/wk,连续8wk后,大鼠血清E2水平显著升高,子宫重量和脏器系数增加。大鼠子宫明显变形,肌层增生,部分大鼠肉眼可见浆膜下肌瘤或肌壁间肌瘤形成。病理组织学检查显示,肌瘤细胞与人子宫肌瘤病理组织学特征相似,肌瘤组织中雌、孕激素受体(ER, PR)表达明显增强。 [结论]大鼠肌肉注射E2200μt/只,2次/wk,连续8wk后可建立与人相似的子宫肌瘤动物模型,适合用于临床发病机制研究和药物筛选。
[Abstract]:Background: uterine leiomyoma is the most common benign tumor in female reproductive system. The data show that estrogen is the main promoting factor of leiomyoma growth. At present, there is a lack of suitable animal models for clinical pathogenesis research and drug screening. The amount and cycle of drug administration in the model is still controversial, which is worth further exploration. Objective to establish an animal model of uterine leiomyoma suitable for clinical pathogenesis research and drug screening. [methods] sexual mature rats were randomly divided into two groups: 200 渭 g and 300 渭 g doses of estrogen (E _ 2). Some animals were killed at 8wk and 10wk. The levels of estrogen (E _ 2) and progesterone (P) in serum were measured, and the changes of uterine morphology were observed. The rate of leiomyoma formation and animal mortality were calculated, and the histopathological characteristics of human uterine leiomyoma were compared. The expression of estrogen and progesterone receptor (ER,PR) in human uterine leiomyoma was also determined. To determine the optimal dose and administration period of E2-induced hysteromyoma formation in rats. [results] after intramuscular injection of E2200 渭 g for 2 times / wk, serum E2 level, uterine weight and organ coefficient increased significantly after continuous 8wk. The uterus was obviously deformed and myometrium proliferated. Subserous myoma or myomyoma could be seen in the naked eyes of some rats. Histopathological examination showed that the histopathological features of leiomyoma cells were similar to those of human uterine leiomyoma. The expression of estrogen and progesterone receptor (ER, PR) was significantly increased in leiomyoma tissues. [conclusion] after intramuscular injection of E2200 渭 t / L / 2 wk, the rat model of uterine leiomyoma similar to human can be established after continuous 8wk, which is suitable for clinical pathogenesis research and drug screening.
