单纯疱疹病毒Ⅰ型糖蛋白B基因疫苗安全性的初步研究

【摘要】: 目的:初步检验单纯疱疹病毒Ⅰ型糖蛋白B基因疫苗对小鼠的安全性。 方法:随机将昆明小鼠分为4组,即对照组、100μg组,300μg组,500μg组,每组6只,雌雄各半。各组分别股四头肌肌肉注射1ml生理盐水、100μg/ml、300μg/ml、500μg/ml pcDNA3-gB,每10天免疫一次,共3次。末次免疫10天后,留取1ml凝血和20ul抗凝血各一份进行血液生理学、血液生化学检测,并对心、肝、脾、肾、脑、角膜、视网膜和三叉神经节8个组织器官进行病理切片检查及基因整合情况的检测。 结果:免疫不同剂量单纯疱疹病毒Ⅰ型糖蛋白B基因疫苗的小鼠全部存活,并且其血液生理学、血液生化学指标、各组织器官的病理切片结果及基因整合情况与对照组小鼠的检测结果未见明显差异(P0.05),不具有统计学意义。 结论:初步说明单纯疱疹病毒Ⅰ型糖蛋白B基因疫苗无病毒致病性,对小鼠是安全的,为下一步应用于大型实验动物及临床奠定了基础。
[Abstract]:Objective: to study the safety of herpes simplex virus type I glycoprotein B gene vaccine in mice. Methods: Kunming mice were randomly divided into four groups: control group, 100 渭 g group, 300 渭 g group and 500 渭 g group. The quadriceps femoris were intramuscularly injected with 1ml saline, 100 渭 g / ml, 300 渭 g / ml, 500 渭 g/ml pcDNA3-gB, once every 10 days, for 3 times. Ten days after the last immunization, blood physiology, blood biochemistry, heart, liver, spleen, kidney, brain, cornea were detected by 1ml coagulation and 20ul anticoagulant. Eight tissues and organs of retina and trigeminal ganglion were examined by pathological section and gene integration. Results: all the mice immunized with different doses of herpes simplex virus glycoprotein B gene vaccine survived, and their blood physiology, blood biochemistry index, The results of pathological sections and gene integration of different tissues and organs were not significantly different from those of control mice (P0.05). Conclusion: herpes simplex virus type I glycoprotein B gene vaccine has no virulence and is safe for mice. It lays a foundation for further application in large laboratory animals and clinic.
