OECs较星型胶质细胞对NSCs分化作用的比较研究

【摘要】: 目的 建立体外纯化培养嗅鞘细胞(Olfactory ensheathing cells, OECs)的稳定方法并观察培养的嗅鞘细胞形态学特征;比较大鼠嗅鞘细胞与星型胶质细胞在体外对神经干细胞(Neural stem cells ,NSCs)分化的影响。 方法 分别从新生SD大鼠的嗅球采用小剂量阿糖胞苷分次纯化法分离、培养嗅鞘细胞;从海马、大脑皮质分离、培养星型胶质细胞、神经干细胞,观察细胞的形态发育特点及免疫组化特性。收集嗅鞘细胞及其上清液、星型胶质细胞及其上清液,将实验分为5组:组Ⅰ:OECs上清液+ NSCs;组Ⅱ: OECs+ NSCs;组Ⅲ:星型胶质细胞上清液+ NSCs;组Ⅳ:星型胶质细胞+ NSCs;组Ⅴ:NSCs单独培养作为对照;将NSCs克隆球平均分成5份,加入各组,不加b-FGF、EFGF、B27,组Ⅰ和组Ⅲ分别用培养了OECs、星形胶质细胞24h的DMEM/F12 ( 1:1 )上清液3ml培养,每24h换液,组Ⅱ和组Ⅳ、组Ⅴ用DMEM/F12 ( 1:1 ) 3ml培养,每24h换液,各组置于相同条件培养箱培养,倒置显微镜下动态观察各组NSCs被诱导分化的细胞的生长情况,并采用免疫组化法对分化细胞鉴定和计数。 结果 1、原代培养后24小时OECs形态基本可辨,背景可见成纤维细胞,第2天开始予小剂量阿糖胞苷分次纯化后成纤维细胞大大减少,OECs的外形主要以突起细长的双极和三极为主,或无突起呈“煎蛋样”,随时间延长细胞生长排列呈现一定方向性;细胞免疫化学示大部分细胞神经生长因子受体P75(neurotrophin receptor p75,NGFRp75)阳性表达。 2、比较大鼠嗅鞘细胞与星型胶质细胞在体外对NSCs分化的影响结果发现:组Ⅰ、组Ⅱ的NSCs球6h已开始贴壁生长,至第7天,多数分化为具有2-3个长突起,胞体圆形或椭圆形的神经元样细胞, NF200 (1:150 )免疫组织化学结果显示这类细胞胞体甚至突触皆被染成棕黄色,胞核不染色,为强阳性,证明该类细胞为神经元,计算其神经元分化率:组Ⅰ为( 51.43±8.58) %,组Ⅱ为(50.11±9.56) % ;组Ⅲ、组Ⅳ大部分细胞NF200 ( 1:150 )染色阴性,GFAP ( 1:150 )阳性,其神经元分化率分别为( 17.25±3.01 ) %、( 15.51±1.62 ) %;组Ⅴ的NSCs球大多数在3-4天后萎缩、漂浮,5-7天基本死亡,神经元分化率为( 0.56±0.33 ) %。经统计学分析嗅鞘细胞组的神经元分化率远远高于星型胶质细胞组及对照组的神经元分化率(P 0.01 )。 3、结论 1、A ra-C抑制法稳定易复制,可获得高纯度的OECs。 2、大鼠嗅鞘细胞(OECs)较星型胶质细胞在体外能更好地促进神经干细胞(NSCs)分化为神经元。
